摘要
优化蒙古黄芪种子的处理方式,进而提高其发芽率。选用磨砂+温水浸种的组合处理,将蒙古黄芪种子的发芽率作为评价指标,以磨砂时间、浸种温度、浸种时间为考察因素,采用单因素试验与响应面分析相结合的方法,优化其处理方式。结果表明,磨砂时间以15 min发芽率最高,达到72%,确定适宜的磨砂时间是10~20 min;浸种温度为60℃时发芽率最高,达到76%,确定适宜的浸种温度是50~70℃;浸种时间以10 min发芽率最高,达到77%,确定适宜的浸种时间5~15 min。各因素对蒙古黄芪种子发芽率影响的大小为:浸种温度>磨砂时间>浸种时间。磨砂时间13 min、浸种温度56℃、浸种时间8 min为蒙古黄芪种子最佳的处理方式,在此处理方式下蒙古黄芪种子的发芽率达90.7%,与预测值92%相近,所优化的处理方式合理、可行,可为生产实践提供参考。
In order to optimize seed treatment of Astragalus mongholicus and improve germination rate,the combined treatment of grinding with sand and soaking seeds with warm water was selected and the germination rate of A.mongholicus seeds were taken as the evaluation index.The grinding time,soaking temperature and soaking time were considered as the indexes,and the single factor test and response surface analysis were used to optimize the treatment.The method of single factor test combined with response surface analysis was used to optimize the treatment.The results showed that the highest germination rate was 72%when the seeds were grinded for 15 min,and the optimum grinding time was 10-20 min.Meanwhile,the highest germination rate reached 76%when the seeds were soaked with 60℃water,and the optimum temperature for seed soaking was 50-70℃.In addition,the highest germination rate was 77%when the seeds were soaked for 10 min,and the optimum soaking time was 5-15 min.To sum up,the influence of various factors on the germination rate of A.mongholicus seeds was as follows soaking temperature>grinding time>soaking time.The optimal treatment were grinding seeds for 13 min and soaking seeds for 8 min at 56℃water,and the germination rate of A.mongholicus seeds reached 90.7%under these treatment methods,which was close to the predicted value of 92%.Therefore,the optimized treatment was reasonable and feasible and could provide reference for production practice.
