摘要
为探讨适合快速检测卵叶海桑花粉活力的方法,采用TTC染色法、MTT染色法、亚历山大染色法、I_(2)-KI染色法、红墨水染色法和离体萌发法对卵叶海桑花粉活力进行了测定。结果表明,卵叶海桑的花粉活力较高,不同方法测定的花粉活力平均值为83.22%~89.35%(TTC染色法除外)。红墨水染色法与离体萌发法的结果,在1%水平上差异极显著。MTT染色法、亚历山大染色法、I_(2)-KI染色法与萌发法的结果差异都不显著。亚历山大染色法和I_(2)-KI染色法能快速、准确地测定卵叶海桑的花粉活力,是判断花粉活力的简便方法,为进行传粉、繁育系统研究和杂交育种提供了理论依据。
In order to explore the suitable method for rapid detection of pollen viability of Sonneratia ovata,TTC staining,MTT staining,Alexander staining,I_(2)-KI staining,red ink staining and in vitro germination method were used to determine the pollen viability of S.ovata.The results showed that the pollen viability of S.ovata was higher,and the average pollen viability measured by different methods ranged from 83.22%to 89.35%(except TTC staining method).The results of red ink staining and in vitro germination were extremely significant at 1%level,while the results of MTT staining,Alexander staining,I_(2)-KI staining and germination method were not significantly different.Alexander staining and I_(2)-KI staining could quickly and accurately determine the pollen viability of S.ovata,which both were simple method to determine the pollen viability,and it could provide a theoretical basis for the study of pollination,breeding and cross breeding.
