人肠道菌群对桃叶珊瑚苷体外转化代谢作用的研究 被引量:2

Effects of human intestinal flora on the metabolic transformation of aucubin in vitro

摘要 目的研究人肠道菌群对桃叶珊瑚苷体外转化代谢的作用。方法采用体外肠道菌孵育方法,在厌氧条件下,将桃叶珊瑚苷分别与3位健康人肠道菌群共同培养48 h,采用高效液相色谱-串联质谱(HPLCMS/MS)联用技术检测不同时间点孵育体系中桃叶珊瑚苷的相对含量及其代谢产物。结果健康人肠道菌群可转化代谢桃叶珊瑚苷,其主要代谢产物为桃叶珊瑚苷苷元和aucubmines A;其主要代谢途径为脱糖基、氨基化及氧化作用。不同受试者肠道菌群对桃叶珊瑚苷代谢速率存在差异,而桃叶珊瑚苷苷元和aucubmines A在共孵育2 h和4 h后可分别被检测到。结论桃叶珊瑚苷可被健康人肠道菌群代谢,4~8 h主要代谢产物为桃叶珊瑚苷苷元,8 h后代谢产物主要为aucubmines A。 Objective To study the effects of human intestinal flora on the metabolic transformation of aucubin in vitro.Methods Intestinal bacteria were incubated in vitro.In anaerobic condition,aucubin was incubated with intestinal flora from 3 healthy people for 48 h.High performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS)was used to determine the relative content and metabolites of aucubin in the incubation system at different point in time.Results The intestinal flora from healthy people metabolized and transformed aucubin,the main metabolites thereof were aucubin aglycone and aucubines A;and the main metabolic pathways thereof were deglycosylation,a mination and oxidation.The metabolic rate of aucubin was different among intestinal flora from diffe-rent subjects,and aucubin and aucubines A could be detected after incubation for 2 h and 4 h,respectively.Conclusion Aucubin can be metabolized by intestinal flora from healthy people,the main metabolite is aucubin aglycone for 4 to 8 h,and the main metabolite is aucubmines A after 8 h.
