褪黑素对内蒙古绒山羊皮肤上皮细胞增殖及wnt10b和β-catenin基因表达的影响 被引量:3

Effects of melatonin on the proliferation of skin epithelial cells and expressions of Wnt10b and β-catenin genes in Inner Mongolian cashmere goats

摘要 为研究褪黑素对内蒙古绒山羊皮肤上皮细胞生长的作用模式,将褪黑素浓度设置为0、100、300、600和900pg/mL 5个试验组,利用CCK-8法检测各组皮肤上皮细胞增殖情况,实时荧光定量PCR检测Wnt10b和β-catenin基因的表达,ELISA检测Wnt10b及β-catenin蛋白的表达。结果表明:300pg/mL褪黑素对皮肤上皮细胞增殖效果最佳(P<0.05),Wnt10b和β-catenin的mRNA和蛋白质表达量极显著高于其他组(P<0.01)。综上,300pg/mL的褪黑素可显著促进皮肤上皮细胞增殖,且显著提高Wnt10b和β-catenin的表达。 In order to study the effect of melatonin on the growth of skin epithelial cells of Inner Mongolia cashmere goats,five test groups were set at 0,100,300,600 and 900 pg/mL of melatonin.CCK-8 method was used to detect the proliferation of skin epithelial cells.Quantitative PCR was to detect the expression of Wnt10 bandβ-catenin gene,and ELISA was used to detect the expression of Wnt10 bandβ-catenin protein.The results showed that 300 pg/mL melatonin had the best effect on the proliferation of skin epithelial cells(P<0.05),and the mRNA and protein expressions of Wnt10 bandβ-catenin were significantly higher than those in other groups(P<0.01).In summary,300 pg/mL melatonin can significantly promote the proliferation of skin epithelial cells and increase the expressions of Wnt10 bandβ-catenin.
