基于生物信息学方法分析PTGIS基因在膀胱癌中的表达及临床意义

摘要 目的:利用Oncomine、GEPIA、LinkedOmics数据库分析前列环素合酶(prostacyclin synthase,PTGIS)在膀胱癌中的表达及与临床预后的关系。方法:应用Oncomine、GEPIA数据库中关于PTGIS研究的数据,对其在膀胱癌中的表达水平变化进行分析。采用GEPIA数据库分析PTGIS表达水平与膀胱癌患者生存预后之间的关系。LinkedOmics数据库分析PTGIS基因表达与膀胱癌临床病理特征的相关性。应用Genecards数据库收集与PTGIS基因有关的蛋白,并通过STRING绘制PTGIS有关蛋白网络图,分析蛋白富集的生理过程。结果:对Oncomine数据库、GEPIA数据库关于膀胱癌和膀胱正常组织中PTGIS基因差异表达的研究,结果显示膀胱癌组织中PTGIS基因的表达显著低于膀胱正常组织(P<0.05)。通过GEPIA数据库生存分析发现PTGIS基因低表达患者的总体生存率明显高于高表达患者,低表达患者的预后更好(P<0.05)。通过Genecards数据库收集到与PTGIS有关的蛋白有CYP51A1、PTGES、PTGDS、AKT1、TBXAS1等30个,其有关蛋白富集分析结果显示主要富集在前列腺素代谢、调节活性氧代谢、炎症反应等生理过程。结论:PTGIS基因在膀胱癌中低表达,其表达水平与生存预后相关,PTGIS基因可为膀胱癌的治疗提供参考,为其进一步深入研究提供理论基础。 Objective:To analyze the relationship between the expression of prostacyclin synthase(PTGIS)in bladder cancer and clinical prognosis using Oncomine,GEPIA,and LinkedOmics databases.Methods:The data of PTGIS research in Oncomine and GEPIA databases were used to analyze the changes of their expression levels in bladder cancer.The relationship between PTGIS expression level and survival prognosis of bladder cancer patients was analyzed using GEPIA database.LinkedOmics database analyzes the correlation between PTGIS gene expression and BLCA clinicopathological characteristics.The Genecards database was used to collect proteins related to the PTGIS gene,and the network map of PTGIS related proteins was drawn through STRING to analyze the physiological process of protein enrichment.Results:The results of Oncomine database and GEPIA database on the differential expression of PTGIS genes in bladder cancer and normal bladder tissues,showed that the expression of PTGIS genes in bladder cancer tissues was significantly lower than that in normal bladder tissues(P<0.05).The survival analysis of GEPIA database found that the overall survival rate of patients with low expression of PTGIS gene was significantly higher than that of patients with high expression,and the prognosis of patients with low expression was better(P<0.05).There were 30 proteins including CYP51A1,PTGES,PTGDS,AKT1,TBXAS1 and other proteins related to PTGIS collected through the Genecards database.The results of protein enrichment analysis showed that they were mainly enriched in the physiological processes of prostaglandin metabolism,regulation of active oxygen metabolism,and inflammation.Conclusion:The PTGIS gene is under-expressed in bladder cancer,and its expression level is related to survival prognosis.The PTGIS gene can provide a reference for the treatment of bladder cancer and provide a basis for its further research.
