摘要
目的:观察Notch1在低氧预适应及DNA甲基转移酶抑制剂5-Aza-cdR注射小鼠海马中的表达变化,探究Notch1在低氧耐受神经保护作用中的可能机制。方法:ICR小鼠通过侧脑室注射5-Aza-cdR及建立低氧预适应模型,通过Western-blot、qRT-PCR及免疫荧光技术,检测小鼠海马Notch1以及DG区神经细胞增殖情况。结果:与未注射5-Aza-cdR组相比,5-Aza-cdR组中Notch1 mRNA表达明显升高(P<0.05);在未注射5-Aza-cdR组中,HPC组较对照组的Notch1的mRNA表达升高(P<0.05),5-Aza-cdR组中5-HPC组较5-C组的Nothc1 mRNA表达升高(P<0.05);相比未注射5-Aza-cdR组,注射5-Aza-cdR各组中Notch1蛋白表达升高(P<0.05),未注射5-Aza-cdR组中HPC组较C组的Notch1蛋白表达升高(P<0.05),5-Aza-cdR组中5-HPC组较5-C组的Notch1蛋白表达增加(P<0.05);相比未注射5-Aza-cdR组,5-Aza-cdR组中海马DG区BrdU阳性细胞数量增多。结论:5-Aza-cdR可以激活Notch1表达,并使海马DG区内神经细胞增殖。
Objective:To observe the expression changes of Notch1 in the hippocampus of mice injected with hypoxic preconditioning and DNA methyltransferase inhibitor 5-Aza,and to explore the possible mechanism of Notch1 in the neuroprotection of hypoxia tolerance.Methods:ICR mice were injected with 5-Aza into the lateral ventricle and a hypoxic preconditioning model was established.Western-blot,qRT-PCR and immunofluorescence techniques were used to detect the proliferation of Notch1 and DG neurons in the mouse hippocampus.Results:Compared with the control group,the expression of Notch1 mRNA in the 5-Aza group was significantly increased(P<0.05).In the control group,the expression of Notch1 in HPC was higher than that in the C(P<0.05).The expression of HPC was higher than that of 5-C Nothc1(P<0.05).Compared with the control group,the expression of Notch1 protein in the 5-Aza group was increased(P<0.05),and the expression of HPC in the control group was significantly higher than that of C(P<0.05).The expression of 5-HPC in the 5-Aza group was significantly higher than that in the 5-Aza group.The expression of C Notch1 increased(P<0.05).Compared with the control group,the number of BrdU-positive cells in the hippocampal DG area increased in the 5-Aza group.
