摘要
目的:观察NgR基因沉默的神经干细胞和雪旺细胞-PLGA支架共移植于创伤性脑损伤大鼠皮层损伤区的运动功能恢复情况。方法:建立大鼠创伤性颅脑损伤模型,分为正常组(Ⅰ组)、单纯PLGA移植组(Ⅱ组)、正常NgR基因的PLGA分子器件移植组(Ⅲ组)、沉默NgR基因的PLGA分子器件移植组(Ⅳ组),每组12只。于皮层损伤区分别植入对应的复合物,并于建模后1、2、3、4周进行改良BBB运动功能评定,并行HE染色观察神经干细胞的存活及分化情况。结果:改良BBB评分Ⅲ、Ⅳ组优于Ⅱ组(P<0.05);Ⅳ组优于Ⅲ组(P<0.05)。建模后1个月,损伤区组织HE染色,Ⅲ组、Ⅳ组移植处的空洞较Ⅱ组少,但是Ⅳ组较Ⅲ组明显。结论:NgR基因沉默的NSC和SC-PLGA支架共移植能够明显促进损伤大鼠的神经元轴突再生,并有助于运动功能的恢复。
Objective:To observe the co-transplantation of NgR gene-silenced neural stem cells and Schwann cell-PLGA scaffold in the cortical injury area of rats with traumatic brain injury to restore motor function.Methods:A model of traumatic brain injury in rats was established and divided into normal group(groupⅠ),simple PLGA transplantation group(groupⅡ),normal NgR gene PLGA molecular device transplantation group(groupⅢ),and NgR gene silencing PLGA molecular device In the transplantation group(GroupⅣ),there were 12 animals in each group.Corresponding compounds were implanted in the cortical injury area,and the modified BBB motor function evaluation was performed 1,2,3,and 4 weeks after modeling.The survival and differentiation of neural stem cells were observed by HE staining.Results:The improved BBB score was better in groupⅢandⅣthan groupⅡ(P<0.05);groupⅣwas better than groupⅢ(P<0.05).One month after modeling,the tissues in the injured area were stained with HE.The graft holes in groupsⅢandⅣwere less than those in groupⅡ,but groupⅣwas more obvious than groupⅢ.Conclusion:Co-transplantation of NgR gene-silenced NSC and SC-PLGA scaffolds can significantly promote neuronal axon regeneration in injured rats,and contribute to the recovery of motor function.
