The Expression of Nectin 4 in breast cancer and its potential role in breast cancer

目的:分析Nectin 4在乳腺癌中的表达水平及其临床意义。方法:采用http://gepia.cancer-pku.cn/网站分析TCGA数据库中乳腺癌患者Nectin 4 mRNA表达水平及其与生存率的关系。采用免疫组织化学法检测62例乳腺癌及59例乳腺良性肿瘤组织中Nectin 4表达水平,采用ELISA法检测乳腺癌、乳腺良性肿瘤患者及健康体检者血清中Nectin 4水平。结果:TCGA数据库中大样本测序结果显示乳腺癌组织中Nectin 4 mRNA表达水平明显高于正常对照组,Nectin 4表达水平与乳腺癌患者生存时间无明显相关性。Nectin 4在乳腺癌组织中表达明显增高,而乳腺良性肿瘤组织中未观察到明显的Nectin 4表达。乳腺癌患者血清Nectin 4为14.54±12.9 ng/L,分别高于乳腺良性肿瘤组的8.65±11.17 ng/L及正常对照组的7.23±6.20 ng/L,差异均有统计学意义(P<0.05),而乳腺良性肿瘤组血清Nectin 4水平与正常对照组比较,差异无统计学意义(P>0.05)。乳腺癌淋巴结转移组血清Nectin 4水平为19.41±14.91 ng/L,高于未转移组的10.09±4.94 ng/L,差异有统计学意义(P<0.05)。血清Nectin 4对乳腺癌诊断的灵敏度为77.4%,特异度为67.8%。结论:乳腺癌组织及血清中Nectin 4高表达,淋巴结转移患者Nectin 4表达更高,提示Nectin 4高表达与乳腺癌的发生发展有关,并促进乳腺癌细胞的浸润及转移,Nectin 4有可能成为乳腺癌诊断和预后监测的潜在分子标志物。

Objective:To investigate the expression and function of Nectin-4 in breast cancer.Methods:Use http://gepia.cancer-pku.cn/website to analyze the Nectin-4 mRNA level in breast cancer patients in TCGA database,and investigate the association between Nectin-4 mRNA levels and overall survival(OS)of breast cancer patients.Immunohistochemistry was used to detect the expression of Nectin-4 in 62 breast cancer tissues and 59 benign breast tumor tissues.Moreover,the serum of 62 cases of breast cancer,59 cases of benign breast tumors and 40 healthy controls were collected,and the level of Nectin-4 was detected by ELISA.Results:The TCGA database of large sample sequencing results showed that the expression level of Nectin-4 mRNA in breast cancer patients was significantly higher than that in the control group.In addition,the survival curve analysis showed that the expression level of Nectin-4 was not significantly correlated with the prognosis of breast cancer.The IHC results indicated that the expression of Nectin-4 is strongly positive in breast cancer tissue.Moreover,the serum level of Nectin-4 in breast cancer patients was significantly higher than that in the benign breast tumor group and the control group(P<0.05).In addition,it was found that serum Nectin-4 levels in stage III/IV breast cancer patients were higher than those in stage I/II patients,and the serum level of Nectin-4 has a positive relationship with lymph node metastasis.Conclusion:The expression level of Nectin-4 is significantly increased in breast cancer patients,and is closely related to breast cancer progression.Nectin-4 might be used as a potential molecular marker for breast cancer diagnosis in the future.
