环状RNA与冠心病的相关性研究

摘要 目的研究新的环状RNA hsacirc0009590、hsacirc0003302、hsacirc0023903与冠心病(CHD)的相关性以及hsacirc0043813基因的单核苷酸多态性位点(SNP)位点rs2293152与心梗风险的相关性。方法提取14例冠心病患者及14例对照组的外周血RNA,采用实时荧光定量PCR检测hsacirc0009590、hsacirc0029303及hsacirc0003302在冠心病及对照人群中的表达差异。提取450例冠心病患者的外周血DNA,采用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)法检测SNP位点rs2293152与冠心病患者的心梗风险的相关性。结果与对照组相比,冠心病患者的hsacirc0009590、hsacirc0003302、hsacirc0029303表达水平均无显著差异(P>0.05),且与血糖、血脂等冠心病危险因素也无明显相关性(P>0.05);而rs2293152与冠心病患者的心梗发生风险无明显相关性(P>0.05)。结论hsacirc0009590、hsacirc0029303、hsacirc0003302与冠心病的患病风险可能无关,SNP位点rs2293152与冠心病的心梗风险可能无关。 Objective To explore the relationship between hsacirc0003302,hsacirc0009590,hsacirc0023903,rs2293152 and coronary heart disease.Methods A total of 14 coronary heart disease(CHD)patients and 14 control patients were recruited,the expression of hsacirc0009590,hsacirc0029303 and hsacirc0003302 were detected by real-time qPCR assay.The relationship between rs2293152 and myocardial infarction(MI)were detected with PCR-restriction fragment length polymorphism(PCR-RFLP)assay.Results There were no significant differences in the expressions of hsacirc0009590,hsacirc0029303 and hsacirc0003302 between CHD and the control group(P>0.05).The genotype distribution or allele frequency of rs2293152 were comparable between MI group and control group(P>0.05).Conclusion hsacirc0009590,hsacirc0029303 and hsacirc0003302 may not correlate with risk of CHD,and SNP site rs2293152 may not correlate with risk of MI in CHD patients.
