摘要
目的探讨子痫前期(PE)患者胎盘组织中microRNA-205-5p(miR-205-5p)的表达情况及其对血管内皮生长因子A(vascular endothelial growth factor A,VEGFA)表达的影响。方法选取2017年12月至2019年6月于河北省人民医院产科住院行剖宫产分娩的PE患者52例(PE组)和正常妊娠孕妇55例(对照组)。荧光定量PCR(RT-qPCR)检测两组患者胎盘组织中miR-205-5p与VEGFA mRNA的表达情况,应用双荧光素酶报告基因实验验证两者的靶向关系,并采用受试者工作特征(ROC)曲线分析胎盘组织中miR-205-5p表达对PE诊断的价值。结果 RT-qPCR结果显示,与对照组相比,PE组胎盘组织中miR-205-5p的表达量显著升高,VEGFA m RNA的相对表达水平显著降低,差异有统计学意义(P<0.05),且胎盘组织中miR-205-5p表达量与VEGFA mRNA表达量呈显著负相关关系(r=-0.652,P<0.05),双荧光素酶报告基因实验证实miR-205-5p与VEGFA存在靶向作用关系。胎盘组织中miR-205-5p表达水平预测PE的ROC曲线下面积为0.933(95%CI 0.8880.978,P<0.05),最佳截断点为1.55,预测PE的灵敏度和特异度分别为84.6%和90.9%。结论胎盘组织中miR-205-5p表达增加可能通过抑制VEGFA的表达在PE发生中起重要作用,且miR-205-5p表达水平对PE的诊断有重要价值。
ObjectiveTo investigate the expression of miR-205-5 p and its effect on vascular endothelial growth factorA(VEGFA)in placenta tissues from patients with preeclampsia.MethodsFifty-two pregnant women withpreeclampsia who had caesarean delivery in the Department of Obstetrics of Hebei Genaral Hospital from December2017 to June 2019 were enrolled as preeclampsia group(PE group),and 55 normal pregnant women undergoingcaesarean section during the same period were selected as control group. RT-q PCR was used to test the expression ofmi R-205-5 p and VEGFA m RNA. Dual Luciferase Reporter Gene Assay was performed to validate the targetingrelationship between them. In addition,ROC curve was used to analyze the assistant diagnostic value of mi R-205-5 pexpression for preeclampsia.ResultsRT-q PCR results showed that the expression levels of mi R-205-5 p weresignificantly higher in PE group(P<0.05),and the relative expression levels of VEGFA m RNA were significantly lowerin PE group(P<0.05)than that in the control group. Moreover,the mi R-205-5 p expression was significantly negativelycorrelated with the m RNA levels of VEGFA(r=-0.652,P<0.05). Dual Luciferase Reporter Gene Assay confirmed thatVEGFA had targeting relationsp with mi R-205-5 p. The results of ROC curve analysis showed that the expression levelof mi R-205-5 p had auxiliary diagnostic value for preeclampsia(P<0.05). The area under the ROC curve was 0.933(95% CI 0.888-0.978;P<0.05),and best cut-off value for preeclampsia was 1.55,the sensitivity being 84.6%,specificity being 90.9%.ConclusionThe increased miR-205-5 p expression in placenta tissue may play an importantrole in the development of PE through inhibiting theexpression of VEGFA,and mi R-205-5 p expression levelcan serve as a predictive indicator for preeclampsia.
