儿童哮喘患者炎症因子水平及肺功能变化与SOCS3基因多态性的相关性 被引量:4

摘要 目的研究SOCS3基因rs9914220和rs4969170位点单核苷酸多态性与儿童哮喘的相关性。方法收集124例哮喘儿童(哮喘组),74例健康对照儿童为对照组,收集患儿的一般临床资料;酶联免疫吸附试验检测外周血中炎症因子白细胞介素(IL)-4、IL-7、IL-10、IL-17和IL-33的水平;检测受试者肺功能指标;全血基因组DNA抽提及检测SOCS3基因rs9914220和rs4969170位点多态性。结果哮喘组男73例,女51例,平均年龄(7.8±2.4)岁,对照组男39例,女35例,平均年龄(7.4±1.8)岁,两组在性别和年龄之间差异无统计学意义(P>0.05)。哮喘组外周血中炎症因子IL-4、IL-7、IL-17和IL-33的水平明显高于对照组,IL-10的水平明显低于对照组(P0.05);对照组和哮喘组rs9914220和rs4969170位点CC和TT基因型差异无统计学意义(P>0.05),CT基因型差异有统计学意义(χ^(2)=5.630,P=0.018;χ^(2)=6.886,P=0.009)。CT基因与哮喘的发生呈负相关,CT基因为预防哮喘发生的保护性因素。结论炎症因子和肺功能的检测有助于儿童哮喘的诊断,SOCS3基因rs9914220和rs4969170位点CT基因型为预防儿童哮喘发生的保护因素。 Objective To investigate the correlations between single nucleotide polymorphisms of SOC993 gene rs9914220 and rs4969170 and asthma in children.Methods The prospective analysis of 124 children with asthma and 74 healthy controls from 2015 to 2018 in our hospital was conducted.The general clinical data of the children were collected.The levels of inflammatory factors IL-4,IL-7,IL-10,IL-17 and IL-33 in peripheral blood were assessed by enzyme-linked immunosorbent assay.The lung function indexes of the subjects were tested.Whole blood genomic DNA extraction was applied to the detection of SOCS3 gene rs9914220 and rs4969170 polymorphisms.Results There were 73 patients in the asthma group(51 females,mean age of 7.8±2.4 years);and 39 healthy controls(35 females,mean age of 7.4±1.8 years).The levels of inflammatory cytokines IL-4,IL-7,IL-17 and IL-33 in the peripheral blood of asthma group were significantly higher than those in the control group;and the level of IL-10 was significantly lower than that in the control group(P0.05).There was no significant differences in CC or TT genotypes in rs9914220 or rs4969170 between the control group and asthma group.The difference in CT genotype was statistically significant(χ^(2)=6.886,P=0.009).The CT gene was significantly negatively correlated with the occurrence of asthma,and the CT gene was a protective factor for asthma.Conclusion The detection of inflammatory factors and lung function is helpful in the diagnosis of asthma in children.The SOCS3 gene rs9914220 and rs4969170 site CT genotypes are protective factors for preventing asthma in children.
