梅花鹿锯茸前后急性期反应血清蛋白含量的变化

摘要 家养梅花鹿每年都要经历1~2次锯茸,锯茸后鹿茸截面可形成直径1~5 cm的创口,其表面不采用任何抗炎药物处理却不引起溃烂及深层感染,并且可以快速愈合,其内在机制目前尚不明确。为探究梅花鹿锯茸后机体炎症变化及肽聚糖与炎症反应的关系,选取9只健康的1岁龄长有毛桃茸公鹿,随机分成3组,分别为空白组(锯茸后创口不做处理)、对照组(锯茸后创口碘伏处理)和实验组(锯茸后创口碘伏+肽聚糖处理),每组3只,于锯茸后的0 d、2 d、4 d、6 d、9 d、11 d、20 d和22 d采集血液,采用双抗体一步夹心法进行酶联免疫吸附实验(Elisa)检测血清中前白蛋白(PA)、转铁蛋白(TRF)、C-反应蛋白(CRP)、血清淀粉样蛋白(SAA)和结合珠蛋白(HP)不同时间点的组内差异及同一时间点的组间差异。结果显示,梅花鹿锯茸后血清中各蛋白含量浓度变化范围较大,个体间差异明显,不同处理方法对同一天急性期5种血清蛋白的含量无显著影响(P>0.05),同一种处理方法不同天数间的5种血清蛋白的含量也无明显差异(P>0.05),但不同个体的部分蛋白在0~6 d变化趋势相似。因此认为家养梅花鹿血清中急性期蛋白存在较大变异范围,锯茸前后血清中急性期蛋白未发现显著变化趋势,机体可能未发生明显的炎症反应,且肽聚糖处理创口未能显著影响血清急性期蛋白水平。 It is found that sika deer has fantastic capacity of anti-infection after the velvet antler was removed,but the mechanismis unclear.To explore the mechanismof this phenomenon,the changes of inflammation in sika deer was evaluated after velvet was removed,the concentration of five kinds of acute phase protein in serumwere determined.In this research,nine male sika deer of the same age with hairy peach antler were selected fromthe breeding farmbelong to the Institute of Special products of the Chinese Academy of Agricultural Sciences,which were randomly divided into a blank group(no treatment),a control group(iodour treatment)and an experimental group(iodoxy+peptidoglycan treatment),three individuals in each group respectively.Blood was collected on the 0th,2nd,4th,6th,9th,11th,20th and 22th day after velvet antler was removed,and prealbumin(PA),transferrin(TRF),C-reactive protein(CRP),serum amyloid A(SAA)and haptoglobin(HP)in serum were detected by enzyme-linked immunosorbent assay(Elisa)using a double antibody one-step sandwich method.The results showed that there were no significant differences in concentration of the five acute phase proteins between different treatment methods and different days(P>0.05).In the population studied,the concentration of the five acute phase proteins had a wide range of values,whose difference between individual were obvious.This result was confirmed by additional determination in larger population of healthy sika deer.Peptidoglycan treatment methods fail to cause significant changes in acute phase proteins,but some proteins are has a similar trend from0 d to 6 d.In conclusion,the concentration of this five acute phase proteins above had a wide range of values in population of healthy sika deer.And the removal of velvet antler may not cause the serious inflammation in sika deer,but further researches need to be done.
