摘要
纸芯片的分析检测技术日趋成熟和多样化,其检测稳定性是确保纸基分析平台准确检测的前提,也是该领域研究的难点之一.前期利用各种微调控手段对纸基材进行改性处理,获得了对于生物标记物吸附量提高的纸芯片产品.本研究通过改变储存时间、储存温度、湿度、环境pH、外来污染物等因素,对微调控手段处理过的纸芯片产品进行葡萄糖氧化酶的检测稳定性研究.选定Ru(bpy)_(3)Cl_(2)·3H_(2)O荧光指示剂,通过监测反应体系荧光强度的变化来表征葡萄糖氧化酶的稳定性.结果表明:PEI与添加木素的微调控手段获得的改性纸芯片产品对于葡萄糖氧化酶的瞬时吸附量相对较佳,与定性滤纸相比分别提高了92%和88%;微调控手段处理后的纸芯片产品在环境温度为25℃、湿度为50%时的储存时间为45~55天,较定性滤纸提高5~10天,其中引入氨基及添加木素手段分别为50天和45天;微调控纸芯片产品对葡萄糖氧化酶的环境温度适用范围为20℃~60℃、环境湿度适用范围为20%~100%、环境pH适用范围为4~7.5,外来污染物(油污、尘土)对载酶纸芯片的酶活性均影响较小;PEI与添加木素的微调控手段获得的改性纸芯片产品对各个影响因素的检测稳定性表现较佳.
The analysis and detection technology of paper chip is becoming more and more mature and diversified.The detection stability of paper chip can support the accurate detection of paper-based analysis platform,and also,it′s the hotpot issue in the paper-based devices.In the early stage,the paper substrate was modified by various microregulatory means,and the paper chip products with higher adsorption capacity for biomarkers were obtained.In this study,detection stability of paper chip was indicated through researching detection stability of GOx immobilized on paper surface by changing the storage time,storage temperature,humidity,environmental pH,foreign pollutants and other factors.Ru(bpy)_(3)Cl_(2)·3H_(2)O fluorescence indicator was selected to characteristic the stability of GOx by monitored the fluorescence intensity changes of reaction system.The results showed that lignin-adding paper and PEI modified paper obtained better GOx instantaneous adsorption,which was 92%and 88%higher than qualitative filter paper.The storage time of micro-control paper chip is 45~55 days under the 25℃and 50%environmental condition humidity,which is 5~10 days higher than filter paper.Meanwhile,in the same environmental condition the storage times of lignin-adding paper and PEI modified paper are 50 and 45 days respectively.The temperature range of micro-control paper chip products that GOx immobilized is 20℃~60℃,environmental humidity is 20%~100%,and pH value is 4~7.5;Foreign pollutants(oil and pollution dust)weakly influenced the activity of immobilization enzyme on the paper chip.Lignin-adding paper and PEI modified paper chip products show better detection stability in various influencing factors.
