摘要
为了探究细粒棘球绦虫来源的miR-71(egr-mi R-71)对绵羊外周血单核细胞(PBMCs)的免疫调节作用,利用egr-mi R-71模拟物和阴性对照分别转染绵羊PBMCs,通过qPCR分析了对绵羊PBMCs中细胞因子和LPS/TLR4信号通路中关键基因表达的影响,并且还利用Griess试剂测定了NO的分泌水平。结果显示,egr-mi R-71的过表达会显著上调CD14,为阴性对照组的2.21倍(P<0.01),但IL-1α、IL-1β、TNF-α的表达则显著下调,分别是阴性对照组的74%、59%和76%(P<0.05)。NO的测定结果还显示,egr-mi R-71能明显促进NO的生成(P<0.05)。以上结果提示,egr-mi R-71对绵羊PBMCs具有免疫调节作用。
The study aims to explore the immunomodulatory effect of Echinococcus granulosusderived mi R-71(egr-mi R-71)on sheep peripheral blood mononuclear cells(PBMCs).PBMCs were transfected with egr-mi R-71 mimics and negative control,respectively.The expression of cytokines and key genes in the LPS/TLR4 signaling pathway in PBMCs was quantitatively analyzed by q PCR,and the NO levels were determined using Griess reagent.The results showed that the overexpression of egr-mi R-71 induced the upregulation of CD14,being 2.21 times compared to the negative control(P<0.01),while IL-1α,IL-1βand TNF-αwere significantly down-regulated by 0.74,0.59 and 0.76 times(P<0.05).In addition,the level of NO was significantly increased in the egr-mi R-71-transfected PBMCs(P<0.05).The results indicate that egr-mi R-71 plays a role in modulation of immune functions of sheep PBMCs.
