人工感染猪繁殖与呼吸综合征病毒猪脑组织的转录组学分析 被引量:2

Transcriptome analysis of porcine brain tissue artificially infected with porcine reproductive and respiratory syndrome virus

摘要 运用Illumina高通量测序技术对感染猪繁殖与呼吸综合征病毒(porcine reproductive and respiratory syndrome virus,PRRSV)的猪脑组织进行RNA-Seq测序,试图分析感染PRRSV的猪脑组织中炎症相关基因以及炎症信号通路的作用。结果显示,PRRSV进入脑组织引起炎症反应,镜检观察发现有大量的淋巴细胞浸润、血管袖套的形成、神经元坏死等非化脓性脑炎的典型病理变化。对转录组测序结果进行分析,发现了一些与炎症紧密相关的基因如TNF-α、CCL2、CXCL9、CXCL10、CCL11等差异性表达,与炎症紧密相关的通路如NF-κB通路、Toll样受体信号通路、TNF信号通路、细胞因子受体相互作用等被激活,提示这些基因和通路均参与了PRRSV感染后脑组织的病理学变化过程。 Illumina high-throughput sequencing technology was used to perform RNA-Seq sequencing on the brain tissues of pigs infected with porcine reproductive and respiratory syndrome virus(PRRSV),analyzing the effects of inflammation-related genes and inflammation signal pathways in the brain tissue of pigs infected with PRRSV.The results found that PRRSV entered the brain tissue to cause inflammatory reactions.Microscopic examination revealed a large number of lymphocytes,the formation of vascular cuffs,neuronal necrosis and other typical pathological changes of non-suppurative encephalitis.The transcriptome sequencing results found some significantly different genes closely related to inflammation such as TNF-α,CCL2,CXCL9,CXCL10,CCL11,etc,pathways closely related to inflammatory such as NF-κB pathway,Toll-like receptor signaling pathway,TNF signaling pathway,cytokine receptor interaction and other pathways.It shows that these genes and pathways are involved in the brain tissue pathological changes after PRRSV infection.
