AQP9基因在不同闭锁进程水牛卵泡颗粒细胞中的表达及其与细胞凋亡关系的研究 被引量:2

摘要 为了解AQP9基因表达及其与水牛颗粒细胞凋亡的关系,初步阐明它在水牛卵泡闭锁中的作用,首先分析了不同闭锁程度水牛卵泡颗粒细胞中AQP9基因和凋亡相关基因的表达情况。通过构建颗粒细胞凋亡模型、过表达AQP9基因的方法分析了它对细胞凋亡相关基因表达的影响。为进一步了解PKC通路与AQP9基因表达的相关性,添加PKC抑制剂处理水牛颗粒细胞,分析细胞凋亡和基因表达情况。上述研究结果表明,AQP9基因主要参与早期卵泡闭锁的过程,AQP9基因可能通过PKC信号通路调节水牛颗粒细胞的凋亡。 The purpose of this study was to investigate the relationship between AQP9 gene expression and granulosa cell apoptosis in buffalo,and to preliminarily understand its role in follicular atresia in buffalo.Firstly,the expression of AQP9 gene and apoptosis-related genes in follicular granulosa cells of buffalo with different atresia degrees were analyzed.The apoptosis model of granulocytes were established,its influence on the apoptosis-related gene expression was analyzed by overexpressing AQP9 gene.In order to further understand the correlation between PKC pathway and AQP9 geneexpression,PKC inhibitor was added to treat buffalo granulocyte cells to analyze cell apoptosis and gene expression.(Conclusion)The results indicated that AQP9 protein is mainly involved in the process of early follicular atresia,and AQP9 protein may regulate the apoptosis of buffalo granulocyte cells through the PKC signaling pathway.
