摘要
目的探究温肺补气通络方(WFBQTL)对转化生长因子(TGF)-β1诱导A549细胞增殖能力及上皮间质转化的影响。方法培养A549细胞,采用TGF-β1刺激A549细胞建立肺腺上皮增殖及间质转化模型,采用甲基偶氮唑蓝(MTT)法检测作用24、48、72 h后细胞增殖率,并拍照记录细胞形态;采用实时定量PCR法(qPCR)、Western印迹检测增殖迁移和上皮间质转化相关蛋白E-钙黏蛋白(Cadherin)、N-Cadherin、Ⅰ型胶原蛋白(CollagenⅠ)、平滑肌肌动蛋白-α(α-SMA)的改变。结果MTT法发现WFBQTL能够降低TGF-β1诱导的A549细胞增殖,细胞形态变化;qPCR和Western印迹法表明,WFBQTL能够上调E-Cadherin表达,下调N-Cadherin、CollagenⅠ和α-SMA表达。结论WFBQTL能够抑制TGF-β1诱导的A549细胞增殖能力和上皮间质转化过程。
Objective To explore the effect of Wenfei Buqi Tongluo(WFBQTL)prescription on the cell proliferation,morphology,and epithelial-mesenchymal transition in the TGF-β1-induced A549 cells.Methods A549 cells were cultured and induced by TGF-β1 to establish a proliferation and epithelial-mesenchymal transition(EMT)model of lung epithelial cells.After the treatment of WFBQTL prescription,cell proliferation was detected by MTT method,and cell morphology was captured.And the proliferation and EMT-associated protein,including E-Cadherin,N-Cadherin,CollagenⅠ,andα-SMA were detected by qPCR and Western blot.Results MTT analysis showed that the WFBQTL prescription inhibited A549 cell proliferation induced by TGF-β1 and could down-regulate the proliferation and migration ability of A549 cells induced by TGF-β1.Compared with control group,TGF-β1 upregulated the protein E-cadherin,N-cadherin,CollagenⅠ,α-SMA,WFBQTL prescription could inhibit the function.Compared with control group,TGF-βupregulated the gene expressions of E-cadherin,N-cadherin,CollagenⅠ,α-SMA,WFBQTL prescription could inhibit the function.Conclusions WFBQTL prescription could inhibit the proliferation and migration of A549 cells induced by TGF-β1.WFBQTL prescription could down-regulate the A549 cells EMT.
