摘要
目的了解江西省鼠形动物中巴尔通体的携带状况和基因型特征,为巴尔通体感染的疾病预防控制提供科学依据。方法2018-2019年在南城县、广信区、横峰县、广丰区、铜鼓县5个鼠传疾病监测项目点,采集130只鼠形动物的肝、脾组织标本进行巴尔通体分离培养,挑取形态疑似巴尔通体的菌落纯培养后提取核酸,PCR检测巴尔通体属特异性基因gltA,阳性菌株鉴定为巴尔通体。对巴尔通体菌株进行全基因组测序,并基于全基因组序列构建聚类树。结果从130只鼠形动物的组织样本中分离巴尔通体14株,携带率为10.77%。黄毛鼠携带率最高(2/6),其次为黑线姬鼠(8/72)、褐家鼠(3/27)、臭鼬鼱(1/12)。基因组序列分析显示14株巴尔通体分为5个不同的分支,分为5种基因型,聚类结果显示在分离地区和宿主类型上没有聚集性。结论江西省鼠形动物中存在巴尔通体携带,携带的宿主类型多,菌株基因组多样性大,存在与人类致病性相关的基因型,存在人类感染的风险。
This study aimed to understand the infection status and genotype characteristics of Bartonella in myomorphs in Jiangxi province.Between 2018 and 2019,liver and spleen tissue samples were collected from 130 rodents in Nancheng,Guangxin,Hengfeng,Guangfeng,and Tonggu counties in Jiangxi Province for isolation of Bartonella.Suspected Bartonella colonies were selected for culture,and nucleic acids were extracted.The Bartonella specific gltA gene was detected by PCR,and the gltA-positive strains were identified as Bartonella.Whole genome sequencing of Bartonella strains was carried out,and a cluster tree was constructed on the basis of whole genome sequences.Fourteen Bartonella strains were isolated from 130 myomorph tissues,and the carriage rate was 10.77%.The carriage rate in Rattus flavipectus was highest(2/6),followed by Apodemus agrarius(8/72),Rattus norvegicus(3/27),and skunk shrew(1/12).Genome sequence analysis showed that 14 Bartonella strains were divided into five different branches,thus suggesting that they comprised five genotypes.The clustering results showed no aggregation in the isolation area and host type.This study revealed that Bartonella carriage exists in many types of myomorphs in Jiangxi Province.The Bartonella isolates showed high genome diversity and some genotypes were associated with human pathogenicity,suggesting a risk of human infection.
