摘要
目的:探讨叉头框蛋白O3a(FOXO3a)在缺氧小鼠大脑组织中的表达。方法:16只SPF级3月龄C57小鼠随机分为对照组(control)和缺氧组(hypoxia)。两组小鼠分别接受常氧浓度环境及低氧环境处理1 h,应用Western Blot法定量检测FOXO3a蛋白的表达水平;应用免疫荧光染色技术检测FOXO3a蛋白在脑组织中的表达分布。结果:与control组比较,应用Western Blot检测,发现缺氧后小鼠脑组织FOXO3a蛋白表达水平上调;免疫荧光检测显示缺氧小鼠的脑皮层、海马区中FOXO3a蛋白表达水平明显升高(P<0.01),其中神经元及星形胶质细胞的FOXO3a表达均明显升高(P<0.01)。结论:缺氧可导致小鼠脑皮层、海马区神经元及星形胶质细胞FOXO3a蛋白表达上调。
Objective:To investigate the expression of FOXO3a protein in the brain tissue of a hypoxic brain injury poisoning stress mouse model.Method:Sixteen 3-month-old C57 mice were divided into two groups:Hypoxic and control group.Mice’s brains were collected at 1 hour after hypoxic treatment.Western Blot was used to detect the protein expression of FOXO3a.Immunofluorescence was used to detect the protein expression of FOXO3a in whole brain,neurons,and astrocyte.Results:Compared with the control group,Western Blot detection showed that the expression level of FOXO3a protein in the brain tissue of mice was up-regulated after hypoxia;and immunofluorescence detection showed that the expression level of FOXO3a protein in the cerebral cortex and hippocampus of hypoxic mice was significantly increased(P<0.01).Among the brain tissues,the expression of FOXO3a protein in neurons and astrocyte was also significantly increased(P<0.01).Conclusions:Hypoxia can cause up-regulation of FOXO3a protein expression in neurons and astrocyte in the cerebral cortex and hippocampus of mice.
