摘要
目的建立能同时测定草香胃康胶囊中阿魏酸、木香烃内酯、去氢木香内酯、红镰霉素-6-O-β-龙胆二糖苷、决明子苷C、橙黄决明素和美决明子素含量的HPLC法。方法色谱柱:Kromasil C 18液相色谱柱(250 mm×4.6 mm,5μm);柱温:25℃;流动相:乙腈-1 mL·L^(-1)甲酸,梯度洗脱;流速:0.8 mL·min^(-1);检测波长:320 nm(检测阿魏酸),225 nm(检测木香烃内酯和去氢木香内酯),284 nm(检测红镰霉素-6-O-β-龙胆二糖苷、决明子苷C、橙黄决明素和美决明子素)。采用SPSS 26.0统计软件对草香胃康胶囊中7种成分的含量进行聚类分析。结果7种成分分别在0.56~14.00、1.89~47.25、2.03~50.75、5.91~147.75、2.27~56.75、1.69~42.25、1.31~32.75μg·mL^(-1)范围内线性关系良好(r≥0.9991);平均回收率分别为97.88%、98.56%、99.16%、100.04%、99.35%、96.98%、98.01%,RSD值分别为1.47%、1.22%、0.94%、0.69%、1.14%、1.03%、0.91%;10批样品聚类分析为2类。结论该方法操作简便、重复性好,可用于草香胃康胶囊中7种成分的含量测定。
Objective To establish an HPLC method for the determination of ferulic acid,costunolide,dehydrocostuslactone,rubrofusarin-6-O-β-gentiobioside,cassiaside C,aurantio-obtusin and obtusifolin in Caoxiang Weikang Capsules simultaneously.Methods The Kromasil C 18(250 mm×4.6 mm,5μm)was used as the chromatographic column with acetonitrile-1 mL·L^(-1) formic acid as the mobile phase in a gradient elution mode.The column temperature was 25℃,and the flow rate was set at 0.8 mL·min^(-1).The detection wavelengths were set at 320 nm for ferulic acid,225 nm for costunolide and dehydrocostuslacton,and 284 nm for rubrofusarin-6-O-β-gentiobioside,cassiaside C,aurantio-obtusin and obtusifolin.The cluster analysis was conducted for the content results of 7 constituents in Caoxiang Weikang Capsules by SPSS 26.0 statistical software.Results The 7 constituents showed good linear relationships within the ranges of 0.56-14.00,1.89-47.25,2.03-50.75,5.91-147.75,2.27-56.75,1.69-42.25 and 1.31-32.75μg·mL^(-1)(r≥0.9991),whose average recoveries were 97.88%,98.56%,99.16%,100.04%,99.35%,96.98%and 98.01%with the RSDs of 1.47%,1.22%,0.94%,0.69%,1.14%,1.03%and 0.91%,respectively.The samples of 10 batches of Caoxiang Weikang Capsules could be divided into 2 categories.Conclusion The method is easy and repeatable,and can be used as a quality control method for the determination of the 7 constituents in Caoxiang Weikang Capsules.
