摘要
目的研究刺五加总苷对快速眼动(REM)睡眠剥夺幼鼠学习记忆的作用。方法将75只幼鼠随机分为对照组(n=25),模型组(n=25)与实验组(n=25)。模型组和实验组均用改良多平台睡眠剥夺法建立REM睡眠剥夺幼鼠模型,对照组为不加任何干预的正常幼鼠。实验组于造模前腹腔内注射刺五加总苷(200 mg·kg^(-1)·d^(-1)),对照组和模型组腹腔注射等量生理盐水,连续干预7 d。睡眠剥夺72 h后,用Morris水迷宫实验测定幼鼠的学习记忆能力;用苏木精-伊红(HE)染色法检测海马组织病理学特点;用羟胺法与硫代巴比妥酸法分别检测血浆超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量;用蛋白质印迹(Wb)法检测海马组织磷酸化Camp反应元件结合蛋白(pCREB)、NMDA受体2B亚基(NR2B)蛋白的表达水平。结果对照组、模型组与实验组幼鼠的逃避潜伏期分别为(21.31±4.65),(62.84±8.79),(30.63±5.94)s;穿越平台次数分别为(8.39±1.57),(1.86±0.79),(5.91±1.06)次;海马组织pCREB蛋白相对表达量分别为0.98±0.02,0.17±0.06,0.83±0.14;NR2B蛋白相对表达量分别为0.86±0.11,0.52±0.08,0.68±0.13,差异均有统计学意义(均P<0.05)。结论刺五加总苷可明显改善REM睡眠剥夺幼鼠的抗氧化能力,同时上调海马组织pCREB、NR2B蛋白表达,进而改善其学习记忆能力。
Objective To investigate the effect total glycosides of Acanthopanax senticosus on learning and memory of of young rats with rapid eye movement(REM)sleep deprivation.Methods A total of 75 young rats were randomly divided into control group(n=25),model group(n=25)and test group(n=25).Model group and test group were used the modified multi-platform sleep deprivation method to establish the REM sleep deprivation young mouse models,control group was normal young rats without any intervention.Test group was intraperitoneally injected with total glycosides of Acanthopanax senticosus(200 mg·kg^(-1)·d^(-1))before the model was made,control group and model group were intraperitoneally injected with the same amount of normal saline at the same time,and continued intervention for 7 d.After 72 h of sleep deprivation,the Morris water maze test was used to determine the learning and memory ability of young rats;hematoxylineosin(HE)staining was used to detect the histopathological features of hippocampus;hydroxylamine method and thiobarbituric acid method were used to detect plasma superoxide dismutase(SOD)activity and malondialdehyde(MDA)content;Western blotting(Wb)method was used to detect the expression levels of phosphorylated camp response element binding protein(pCREB)and NMDA receptor 2 B subunit(NR2 B)protein in hippocampus.Results The escape latency of control group,model group and test group were(21.31±4.65),(62.84±8.79),(30.63±5.94)s;the number of crossing platforms were(8.39±1.57),(1.86±0.79),(5.91±1.06)times·min-1;the relative expression levels of pCREB protein in hippocampus were 0.98±0.02,0.17±0.06,0.83±0.14;the relative expression levels of NR2 B protein were 0.86±0.11,0.52±0.08,0.68±0.13,all with significant difference(all P<0.05).Conclusion Total glycosides of Acanthopanax senticosus can significantly improve the learning and memory abilities of REM sleep deprivation young rats,and improve its antioxidant capacity,its mechanism may be related to up-regulating the expression of p CREB and NR2 B protein in hippocampus.
