摘要
目的:观察少腹逐瘀汤对高脂血症模型大鼠血小板生物学指标的影响,从血小板变化上探讨少腹逐瘀汤的作用靶点。方法:选取50只SD大鼠为研究对象,随机分为空白组,模型组,辛伐他汀组(0.004g/kg),少腹逐瘀汤高、低剂量组(4.8、1.2g/kg),每组10只。除空白组外,其余各组按大鼠体质量灌胃脂肪乳剂复制高脂血症模型,并在造模第21天开始灌胃给药20d。给药期间,造模各组持续灌胃高脂肪乳剂。给药结束后腹主动脉取血,保存肝脏,检测与血小板生物学活性有关指标以及脂代谢相关指标。结果:与模型组比较,少腹逐瘀汤高剂量组能够显著降低血浆TG、LDL-C含量(P<0.05),显著降低TXA2、TXB2、β-TG、FN、vWF、FIB、PAI-1、TT、PLC、PLA2含量(P<0.05,P<0.01),显著升高PGI2、6-keto-PGF1α含量(P<0.05);少腹逐瘀汤高、低剂量组均能显著提升肝组织中MTP和LPL含量(P<0.05)。结论:少腹逐瘀汤具有降血脂、抑制血小板生理活性的作用。
Objective: To observe the effects of Shaofu Zhuyu Decoction(SFZYD) on platelet biological indexes of hyperlipidemia model rats and to explore the target of SFZYD from the changes of platelet. Methods: Fifty SD rats were randomly divided into blank control group, hyperlipidemia model group, simvastatin group(0.004 g/kg), SFZYD high, low dose group(4.8, 1.2 g/kg) with 10 rats in each group. Except the blank control group, the other groups were given SFZYD for 20 days by intragastric administration of fat emulsion according to the weight of rats to replicate hyperlipidemia model. During the administration period, high fat emulsions were continuously administered to the model groups. After the last administration, blood was taken from the abdominal aort and the indexes related to platelet biological activity were detected. The liver was preserved and lipid metabolism were detected. Results: The content of TG, LDL-C, TXA2, TXB2, β-TG, FN, vWF, FIB, PAI-1, TT, PLC and PLA2 could be reduced significantly by SFZYD high dose group(P<0.05, P<0.01). The content of PGI2、6-keto-PGF1α could be induced significantly(P<0.05). SFZYD high and low dose groups could increase the content of MTP and LPL in the liver(P<0.05). Conclusion: SFZYD has the effect of lowering blood lipid and inhibiting platelet physiological activity.
