摘要
目的通过建立H_(22)移植瘤小鼠模型,探讨鲜人参膏(Fresh ginseng paste,FGP)对H_(22)荷瘤小鼠的抗肿瘤作用。方法将ICR小鼠随机分成模型组、阳性对照组(环磷酰胺,CTX)、FGP低剂量组(300 mg/kg)和高剂量组(600 mg/kg),连续给药15d后,计算抑瘤率和脏器指数,检测血清中谷丙转氨酶(ALT)和谷草转氨酶(AST)水平;并通过酶联免疫试剂盒测定小鼠血清中肿瘤坏死因子-α(TNF-α)和干扰素-γ(IFN-γ)的含量。结果FGP能够明显改善H_(22)荷瘤小鼠的一般状况,抑制肿瘤生长,显著降低脾脏指数及胸腺指数(P<0.05);此外,与模型组相比,FGP能够明显改善肝功能的损伤,并显著升高TNF-α和IFN-γ的水平(P<0.05)。结论FGP对H_(22)荷瘤小鼠具有一定的抗肿瘤作用,并能够减少肝损伤,其作用机制可能与增强机体免疫力有关。
Objective:To establish a mouse model of H_(22)transplanted tumor to explore the anti-tumor effect of fresh ginseng paste(FGP)on H_(22)tumor-bearing mice.Methods:ICR mice were randomly divided into model group,positive control group(cyclophosphamide,CTX),FGP low-dose group(300 mg/kg)and FGP high-dose group(600 mg/kg),after 15 days of continuous administration,calculate the tumor inhibition rate and organ index,and detect the levels of alanine aminotransferase(ALT)and aspartate aminotransferase(AST)in serum;and determine the tumor necrosis factor in mouse serum by enzyme-linked immunoassay kit-α(TNF-α)and interferon-γ(IFN-γ)content.Results:FGP can significantly improve the general condition of H_(22)tumor-bearing mice,inhibit tumor growth,and significantly reduce the spleen index and thymus index(P<0.05);in addition,compared with the model group,FGP can significantly improve liver function damage,and Significantly increased the levels of TNF-αand IFN-γ(P<0.05),and showed a dose-effect relationship.Conclusion:FGP has a certain anti-tumor effect on H_(22)tumor-bearing mice,and can reduce liver damage.Its mechanism of action may be related to the enhancement of the body's immunity.
