阳江市新型冠状病毒肺炎患者核酸检测结果与病程关系的探讨 被引量:4

摘要 目的:建立荧光定量PCR检测新型冠状病毒核酸相对定量的方法,分析COVID-19肺炎患者核酸检测结果与病程的关系。方法:使用荧光定量PCR法对阳江市2020年1月—2020年8月新型冠状病毒肺炎患者进行核酸检测,并对其结果进行分析,确定新冠病毒载量与Ct值之间的相关性,探讨患者样本CT值与病程的关系。结果:该荧光定量PCR法ORF1ab基因与N基因的Ct值与RNA模板含量呈线性负相关。确诊病例第1、3、5周平均Ct值逐渐增大,差异具有统计学意义(P<0.05),患者体内新型冠状病毒呈下降趋势;重型病例第一周平均Ct值比轻型病例第一周平均Ct值低,重型病例核酸平均转阴时间长于轻型病例核酸转阴时间。结论:新型冠状病毒肺炎患者核酸检测Ct值与病毒载量存在相关性,Ct值可辅助监控患者病情发展和病程长短,为医生对病例病程研判提供参考依据。 Objective:To establish a fluorescence quantitative PCR method to detect the relative quantification of nucleic acid with novel Coronavirus 2019 pneumonia and analyze the relationship between the nucleic acid test results and the course of disease in patients with novel Coronavirus 2019 pneumonia.Methods:The nucleic acid test was performed on COVID-19 patients from January 2020 to August 2020 in Yangjiang city by using the fluorescence quantitative PCR method.The results were analyzed to determine the correlation between novel Coronavirus load and Ct value,and to explore the relationship between Ct value and the course of disease in patient samples.Results:Ct values of ORF1ab and N genes were negatively correlated with RNA template content.The mean Ct value of the confirmed cases gradually increased at week 1,3 and 5,and the difference was statistically significant(<0.05).Novel Coronavirus showed a downward trend in the patients.The mean Ct value in the first week of severe cases was lower than that in the first week of mild cases,and the mean time of nucleic acid turning negative in severe cases was longer than that in mild cases.Conclusion:Ct value of NUCLEic acid test in COVID-19 patients is correlated with viral load.Ct value can assist to monitor the development and duration of the patient's disease and provide reference for doctors to study and judge the course of the case.
