摘要
目的:观察围刺对张力性小鼠模型术后增生性瘢痕的修复作用。方法:选取4周龄雌性清洁级小鼠90只,使用机械张力诱发增生性瘢痕。造模完成后将小鼠随机分为空白组(模型组)、对照组(曲安奈德治疗组)和观察组(围刺治疗组)。治疗结束后处死小鼠,行HE染色和Masson染色,VEGF和Ⅰ型胶原蛋白免疫荧光检测等。结果:成功采用机械张力诱发增生性瘢痕模型。观察组小鼠瘢痕组织修复效果与对照组相当,真皮层厚度显著低于空白组(P<0.05),两组比较,差异无统计学意义(P>0.05)。观察组小鼠瘢痕组织中的I型胶原蛋白表达水平均高于空白组(P<0.05),且观察组和对照组比较,差异无统计学意义(P>0.05)。3组小鼠VEGF水平比较,差异无统计学意义(P>0.05)。结论:围刺对张力性小鼠模型术后增生性瘢痕修复作用显著,其作用可能与MAPK信号通路的激活有关。
Objective:To investigate the effect of circumpuncture on repair of postoperative hypertrophic scar in a mouse model of tension.Methods:Ninety four-week-old female clean mice were included.Mechanical tension was used to induce model of hypertrophic scars.After modeling,the mice were randomly divided into the blank group(model group),control group(given triamcinolone acetonide)and study group(receiving circumpuncture).After treatment,the mice were sacrificed.The scar tissues were studied with HE and Masson staining.The expression of VEGF and type I collagen in the scars were detected by immunofluorescence.Results:The hypertrophic scar model was successfully estabished by mechanical tension induction.The study group showed comparable outcome of scar tissue repair compared with the control group,and significantly lower thickness of the dermis compared with the blank group(P<0.05).There was no significant difference in the thickness of the dermis between the study and control groups(P>0.05).The expression of type I collagen in the scar tissue of the study group was higher than that in the blank group(P<0.05),and but there was no significant difference between the study group and the control group(P>0.05).The expression of VEGF did not differ across the three groups of mice(P>0.05).Conclusion:Circumpuncture results in significant effect on repair of hypertrophic scars in mouse model of tension induction.Such effect may be related to activation of MAPK signaling pathway.
