摘要
【目的】研究柚皮素(Naringenin,NAR)对人永生化角质形成细胞(HaCaT cells)氧化损伤的作用。【方法】用MTT检测细胞活性;H&E染色观察柚皮素处理对氧化损伤HaCaT细胞形态的影响;划痕检测细胞迁移能力;克隆检测细胞增殖;蛋白免疫印迹检测蛋白表达量;氧化试剂盒检测SOD和MDA水平。【结果】柚皮素对HaCaT细胞的活性无明显的抑制作用,经柚皮素干预后H2O2损伤的HaCaT细胞迁移加快;柚皮素作用于H2O2损伤的HaCaT细胞后SOD2、CDK2蛋白表达水平上升,细胞SOD活性提高和MDA的含量减少。【结论】柚皮素能恢复H2O2损伤的HaCaT细胞的增殖、迁移,并且可以影响SOD2、CDK2蛋白的表达,提高SOD活性,减少MDA的含量,后续可能发展成为延缓衰老的潜在保护皮肤用药。
【Objective】The aim of this paper is to study the effect of Naringenin(NAR)on human immortalized HaCaT cells and its antioxidant activity.【Method】Cell activity was detected by MTT.H&E staining was used to observe the effect of naringin treatment on the morphology of oxidative damage HaCaT cells.Cell migration ability was detected by scratches.Cell proliferation was detected by cloning.Western blot was used to detect the protein expression.SOD and MDA levels were measured with the oxidation kit.【Result】Naringenin had no obvious inhibitory effect on the activity of HaCaT cells,and the migration of HaCaT cells damaged by H2O2 was accelerated after the intervention of naringienin.After the treatment of naringenin on HaCaT cells damaged by H2O2,the expression level of SOD and CDK protein increased,the activity of SOD increased and the content of MDA decreased.【Conclusion】Naringenin can restore the proliferation and migration of HaCaT cells damaged by H2O2,affect the expression of SOD2 and CDK2 protein,improve SOD activity and reduce the content of MDA.Subsequently,it may be developed into a potential skin protective drug to delay aging.
