准噶尔乌头的生物碱成分及其对一氧化氮生成的抑制作用研究 被引量:1

摘要 目的:研究准噶尔乌头Aconitum soongaricum Stapf的生物碱成分及其对一氧化氮(NO)生成的抑制作用。方法:采用硅胶柱、MCI-gel及Sephadex LH-20凝胶柱层析等多种色谱分离方法,对准噶尔乌头的总生物碱部位进行分离纯化,根据^(1)H-NMR、^(13)C-NMR等波谱数据对所得化合物进行结构鉴定,并评价这些化合物对脂多糖(LPS)诱导的小鼠巨噬细胞NO生成的抑制作用。结果:从准噶尔乌头中共分离得到16个生物碱成分,分别鉴定为:宋果灵(1)、准噶尔乌头胺(2)、azitine(3)、新乌宁碱(4)、欧乌头碱(5)、12-表-脱氢欧乌碱(6)、14-O-乙酰尼奥林(7)、苯甲酰乌头宁(8)、15α-羟基尼奥林(9)、欧乌头碱氮-氧化物(10)、(+)-isoboldine(11)、(+)-glaucine(12)、(+)-N-methyllaurotetanine(13)、N-methyl hordenine(14)、hordenine(15)、N-acetyl hordenine(16)。检测化合物1~13在25μmol/L浓度下对小鼠巨噬细胞NO生成的抑制作用,化合物3的抑制率为(68.2±5.9)%,其余化合物的抑制率均低于3%。结论:其中,化合物3、8、11~16为首次从该植物中分离得到,化合物3具一定的抗炎活性。 Objective:To study the alkaloids compositions of Aconitum soongaricum and the inhibition effect on nitric oxide(NO)production.Methods:The whole alkaloids extraction of Aconitum soongaricum was isolated and purified by silica,MCI-gel and Sephadex LH-20,the structures of obtained compounds were identified by ^(1)H-NMR and ^(13)C-NMR spectral data,and the inhibition effects of these compounds on LPS-induced NO production in mouse macrophages were evaluated.Results:Sixteen alkaloids were isolated from Aconitum soongaricum and identified as songorine(1),songoramine(2),azitine(3),neoline(4),napelline(5),12-epi-dehydronapelline(6),14-O-acetylneoline(7),benzoylaconine(8),15α-hydroxyneoline(9),napelline N-oxide(10),(+)-isoboldine(11),(+)-glaucine(12),(+)-N-methyllaurotetanine(13),N-methyl hordenine(14),hordenine(15),N-acetyl hordenine(16),respectively.The inhibition effect on NO production of compounds 1 to 13 at 25μmol/L concentration were evaluated,the inhibition ratio of compound 3 was(68.2±5.9)%,the other compounds were less than 3%.Conclusion:Among them,compounds 3,8,11 to 16 are isolated from this plant for the first time,and compound 3 has certain anti-inflammatory activity.
