指纹图谱结合一测多评法评价杜仲叶代替杜仲皮的可行性 被引量:15

Evaluation of the Possibility of Eucommia ulmoides Leaves Replacing its Barks by Fingerprint Combined with Multi-components Quantitive by One Marker Method

摘要 目的:建立杜仲皮及杜仲叶指纹图谱,结合一测多评(QAMS)法对杜仲叶代替杜仲皮的可行性进行分析。方法:采用YMC-Pack ODS-A C_(18)(150 mm×4.6 mm,5μm)色谱柱;以甲醇-0.5%磷酸溶液为流动相,梯度洗脱;柱温为35℃;流速为1.0 mL/min;检测波长为230 nm;建立杜仲皮、叶指纹图谱,采用主成分分析及正交偏最小二乘判别分析法进行整体观察和评价;采用一测多评法对其中京尼平苷酸、绿原酸、京尼平苷、京尼平、松脂醇二葡萄糖苷进行含量测定。结果:建立了杜仲皮、叶指纹图谱,杜仲皮含有16个共有峰,杜仲叶含有20个共有峰,其中皮与叶间含有6个共有峰,确证与其差异相关的1个成分为5号京尼平苷。采用一测多评法对其中5种成分进行含量测定,结果显示与杜仲皮相比,绿原酸在杜仲叶中含量偏高,京尼平苷酸、京尼平苷及松脂醇二葡萄糖苷在杜仲叶中的含量偏低(P<0.05)。结论:在降血脂、降糖、抗菌消炎等方面杜仲叶或可代替杜仲皮使用。 Objective:To establish the HPLC fingerprints of Eucommia ulmoides barks and its leaves,to analyze the possibility of Eucommia ulmoides leaves replacing its barks by multi-components quantitive by one marker(QAMS)method.Methods:The fingerprints of Eucommia ulmoides barks and Eucommia ulmoides leaves were established by using YMC-Pack ODS-A C_(18)(150 mm×4.6 mm,5μm)column,the mobile phase was methanol-0.5%phosphoric acid solution with gradient elution,column temperature was 35℃,flow rate was 1.0 mL/min and detection wavelength was 230 nm.Principal component analysis and orthogonal partial least square discriminant analysis were used for overall observation and evaluation.The contents of geniposidic acid,chlorogenic acid,geniposide,genipin,pinoresinol diglucoside were determined by QAMS method.Results:The fingerprints of Eucommia ulmoides barks and Eucommia ulmoides leaves were established.There were sixteen common peaks in the barks of Eucommia ulmoides,twenty common peaks in the leaves of Eucommia ulmoides,six common peaks between the barks and leaves.The chromatographic peak of NO.5 geniposide was confirmed to be related to its difference.The contents of five components were determined by QAMS method,and the results showed that compared with the barks of Eucommia ulmoides,the content of chlorogenic acid in the leaves of Eucommia ulmoides was higher,while the contents of geniposidic acid,geniposide and pinoresinol diglucoside in the leaves of Eucommia ulmoides were lower(P<0.05).Conclusion:The Eucommia ulmoides leaves may be a substitute of Eucommia ulmoides barks in lowering blood fat,lowering blood sugar,fighting bacteria and reducing inflammation.
