神术散治疗新型冠状病毒肺炎的分子机制研究 被引量:9

摘要 目的:运用网络药理学研究方法探讨神术散治疗新型冠状病毒肺炎的作用机制。方法:通过中药系统药理学数据库(TCMSP)对神术散所包含的中药进行可能的活性成分和靶点预测,通过Cytoscape 3.7.2建立药物-成分-靶点作用网络,并使用STRING网站进行靶点蛋白互作分析、运用Cytoscape 3.7.2进行可视化;利用DAVID数据库进行基因本体(GO)分析和京都基因与基因组百科全书(KEGG)通路富集分析。结果:筛选出36个神术散的有效成分和125个预测靶点。富集分析共得到生物学进程23个,细胞学组分8个,分子功能9个,11条KEGG通路。结果显示神术散作用于新型冠状病毒肺炎的机制中,其主要为癌症相关通路、PI3K-Akt信号通路、钙离子信号通路、TNF信号通路和HIF-1信号通路。结论:神术散治疗新型冠状病毒肺炎可能存在多通路、多靶点,这为进一步深入揭示其作用机制提供了参考,也为新型冠状病毒肺炎的治疗提供了生物信息学基础。 Objective: To explore the mechanism of Shenzhu powder in the treatment of novel coronavirus pneumonia using network pharmacology.Methods: The possible active components and targets of traditional Chinese medicine contained in Shenzhu powder were predicted by TCMSP database, the drug-component-target interaction network was established by Cytoscape 3.7.2.The target protein interaction was analyzed by STRING website and visualized by Cytoscape 3.7.2.DAVID database was used for gene ontology(GO)analysis and Kyoto encyclopedia of genes and genomes(KEGG)pathway enrichment analysis.Results: 36 active components and 125 predictive targets of Shenzhu powder were selected.A total of 23 biological processes, 8 cellular components, 9 molecular functions and 11 KEGG pathways were obtained by enrichment analysis.The results showed that the mechanism of Shenzhu powder acted on novel coronavirus pneumonia was mainly cancer-related pathway, PI3 K-Akt signal pathway, calcium signal pathway, TNF signal pathway and HIF-1 signal pathway.Conclusion: Shenzhu powder may have multiple pathways and multiple targets in the treatment of novel coronavirus pneumonia, which provides a reference for further revealing its mechanism of action and a bioinformatics basis for the treatment of novel coronavirus pneumonia.
