埃及伊蚊体内wAlbB株沃尔巴克氏体感染的时空分布 被引量:2

The temporal and spatial distribution of the density of infection with the Wolbachia strain wAlbB in Aedes aegypti

摘要 目的了解沃尔巴克氏体(Wolbachia)在WB株埃及伊蚊体内不同时间和空间的分布状况。方法采用实时荧光定量PCR测定埃及伊蚊体内不同发育时期、不同日龄和不同组织的Wolbachia密度。结果埃及伊蚊卵和成虫期Wolbachia密度均高于其幼虫和蛹期(P<0.01);不同日龄雌、雄成蚊体内Wolbachia密度均随日龄增大而上升(P<0.05);同一日龄雌、雄成蚊体内Wolbachia密度随着日龄增长雌蚊显著高于雄蚊(P<0.01);雌蚊卵巢中Wolbachia密度显著高于脂肪体和中肠,但是与唾液腺之间的差异不具统计学意义(P>0.05)。结论wAlbB株Wolbachia密度受埃及伊蚊发育状态和日龄的影响,随日龄升高密度显著升高。这为进一步揭示Wolbachia与宿主之间的互作关系提供了基础数据。 Objective To determine the spatiotemporal distribution of the density of infection with the Wolbachia strain wAlbB in Aedes aegypti.Methods Specific primers were designed based on the sequence of a Wolbachia surface protein gene(wsp)to construct a standard plasmid.The density of infection with Wolbachia was determined during different developmental stages,ages,and body parts in Ae.aegypti using real-time quantitative PCR.Results The density of Wolbachia in eggs and adults was higher than that in the larval and pupal stages.The difference in rates was significant(P<0.01).The density of infection with Wolbachia increased significantly with age in female and male adults(P<0.05).Overall,the prevalence of Wolbachia in female adults was higher than that in males of the same age.The difference in rates was significant(P<0.01).Th highest Wolbachia load was in the ovaries of female mosquitoes,but the load in the ovaries and salivary glands did not differ significantly(P>0.05).Conclusion The density of infection with Wolbachia was affected by the developmental status and age of Ae.aegypti mosquitoes,and Ae.aegypti also has tissue tropism.This study provides basic data revealing interaction between Wolbachia and a host.
