摘要 组织器官纤维化是多种细胞因子、多条信号通路共同作用的产物,转化生长因子β1(TGF-β1)几乎参与了纤维化发生进展的全过程,是当前最强促纤维化细胞因子之一。TGF-β1主要通过TGF-β1/Smads信号通路发挥促纤维化作用,近年来关于该信号通路的研究成果不断增多。中医药因其成分复杂、靶点多样、不良反应少等优势被广泛应用于临床相关疾病的防治。中药复方、中药单体化合物的分子生物学研究也在如火如荼地开展,通过借助现代科学技术手段阐释中药的具体作用靶点及信号通路调控方式,可进一步明确中药疗效机理,对于中医药的科普与推广具有促进推动作用。该文阐述了TGF-β1/Smads信号通路的结构及其对纤维化的调控机制,并系统总结了近年来中医药通过干预TGF-β1/Smads信号通路改善、延缓多种脏器及组织纤维化的具体作用方式和可能机制,为今后纤维化性疾病的中医药靶向精准治疗提供参考。 Fibrosis of tissues and organs is the product of multiple cytokines and multiple signaling pathways.Transforming growth factorβ1 is almost involved in the entire process of fibrosis progression,and is one of the strongest cytokines to promote fibrosis.TGF-β1 mainly plays a role in promoting fibrosis through the TGF-β1/Smads signaling pathway,and the research achievements on this signaling pathway have been increasing in recent years.TCM is widely used in the prevention and treatment of clinical related diseases due to its advantages of complex ingredients,diverse targets,and few side effects.The molecular biology research of TCM compound and monomer compound is also in full swing.By using modern scientific and technological means to explain the specific action targets of TCM and the control methods of signal pathways,the mechanism of the efficacy of TCM can be further clarified,which can promote the popularization and popularization of Chinese medicine.This article describes the structure of TGF-β1/Smads signaling pathway and its regulation mechanism on fibrosis,and systematically summarizes the specific role of TCM in improving and delaying various organ fibrosis by intervening TGF-β1/Smads signaling pathway in recent years,which would provide a reference for future targeted and precise treatment of Chinese medicine for fibrotic diseases.
