活血解毒方治疗糖尿病视网膜病和周围神经病变的谱效关系研究

摘要 目的观察活血解毒方治疗糖尿病视网膜病及周围神经病变的谱效关系,探究活血解毒方的药效物质基础。方法建立活血解毒方的HPLC指纹图谱,制备糖尿病大鼠模型,测定给药后大鼠的血糖、运动神经传导速度与视网膜中央动脉血流速度,采用相关分析与回归分析评价活血解毒方的主要药效成分。结果活血解毒方显著增加视网膜中央动脉血流速度,加快运动神经传导速度。活血解毒方主要贡献成分来自三七和黄连,其中三七总皂苷和小檗碱与糖尿病视网膜病和周围神经病变重要相关。结论活血解毒方可显著改善糖尿病大鼠视网膜病和周围神经病变,且三七和黄连起主要作用。 Objective To observe the spectrum-effect relationship of Huoxue Jiedu Recipe(HXJD)(活血解毒方)on diabetic retinopathy(DR)and diabetic peripheral neuropathy(DPN)and explore the material basis of HXJD.Methods HPLC fingerprint of HXJD was established and the rat model of diabetes was constructed.After 8 weeks of HXJD administration,the blood glucose,motor nerve conduction velocity(MNCV)and blood flow velocity in central retinal artery of the rats were measured.The main effective ingredients of HXJD were calculated by correlation analysis and regression analysis.Results HXJD sped up MNCV and blood flow velocity in retinal central artery.The important contributing components of HXJD came from Panax notoginseng and Coptis chinensis,among which the total saponins of P.notoginseng and berberine were related importantly.Conclusion HXJD significantly improved DR and DPN in diabetic rats.P.notoginseng and C.chinensis were the main contributing components.
