电针频率对脑梗死大鼠缺血区UCP-5表达的影响 被引量:7

摘要 目的目的:通过观察不同频率电针对脑梗死大鼠脑内缺血区的作用,根据UCP-5的表达,探讨电针治疗脑梗死的作用机制。方法:将健康成年雄性SD大鼠随机分为空白组、假手术组、模型组和2、50、100 Hz电针组。电针组取大鼠患侧"前三里穴"和"外关穴"。各组均在术后第1 d、3 d、7 d、14 d、21 d用网屏试验进行评分,对大鼠的神经缺损情况进行评价。采用免疫组织化学的方法,检测其脑缺血区UCP-5的阳性表达。结果:与2Hz组比较,造模后21 d,2 Hz组大鼠的行为学评价优于模型组大鼠,差异有统计学意义(P<0.05)。与50Hz组比较,造模后7d,50 Hz组大鼠行为学评分优于模型组,差异存在统计学意义(P<0.01);造模后14、21 d,50 Hz组大鼠行为学评分优于其他各组,差异有统计学意义(P<0.05)。造模后,大鼠UCP-5阳性表达有所上升。模型组大鼠产生的脑保护作用优于空白组,差异有统计学意义(P<0.01)。不同频率电针治疗后,大鼠UCP-5阳性表达有所上升。与2 Hz组比较,2 Hz组大鼠脑保护作用优于空白组、100 Hz组,差异有统计学意义(P<0.05)。与50Hz组比较:50Hz组大鼠脑保护作用优于空白组,差异具有统计学意义(P<0.01)。结论:不同频率电针可使脑梗死大鼠运动神经功能障碍得到改善,其中2Hz电针组效果最显著,其次为50Hz和100Hz电针组。电针治疗脑梗死对脑具有保护作用,其治疗的机制可能与脑梗死区UCP-5的调节有关。 Objective The effects of electroacupuncture on cerebral infarction were investigated according to the expression of UCP-5 by observing the effects of different frequencies on the ischemic brain in rats with cerebral infarction.Methods Healthy adult male Sprague-Dawley rats were randomly divided into blank group,sham-operated group,model group and 2,50,100 Hz electroacupuncture groups.The electroacupuncture group was used to take the"Qiansanli"(EI10)and"Waiguan"(SJ5)on the affected side of the rats.Each group was scored by screen experiment on the 1 st,3 rd,7 th,14 th and 21 st day postoperatively to evaluate the nerve defect of rats.Immunohistochemistry was used to detect the positive expression of UCP-5 in cerebral ischemia.Results The behavioral evaluation of rats in the 2 Hz group was better than that in the model group with statistical significance(P<0.05).The behavioral evaluation of rats in the 50 Hz group was better than that in the model group 7 days after operation with statistical significance(P<0.01).At 14 and 21 days after model establishment,the behavioral evaluation of rats in the 50 Hz group was better than those of the other groups with statistical significance(P<0.05).The positive expression of UCP-5 increased after the establishment of the model.The difference between the model group and the blank group was statistically significant(P<0.01).The positive expression of UCP-5 in rats increased after electroacupuncture at different frequencies.The protective effect of 2 Hz group was better than that of blank group and 100 Hz group(P<0.05).The protective effect of the 50 Hz group was better than that of the blank group(P<0.01).Conclusion The motor nerve dysfunction of rats with cerebral infarction can be improved by electroacupuncture at different frequencies.The effect of 2 Hz electroacupuncture group was the most significant,followed by 50 Hz and 100 Hz electroacupuncture group.Electroacupuncture has the effect of brain protection in the treatment of cerebral infarction,and the mechanism of its treatment may be related to the up-regulation of UCP-5 in the cerebral infarction area.
