电针调控SIRT1对肥胖大鼠肝脏炎症因子IL-6、TNF-α的影响 被引量:3

摘要 目的:观察电针通过调控SIRT1通路对胰岛素抵抗肥胖大鼠肝脏炎症的影响,探讨电针改善大鼠肥胖状态的机制。方法:将70只Wistar雄性大鼠适应性喂养1周后,随机挑选10只大鼠给予标准饮食喂养,8周后随机抽取6只作为正常组。其余大鼠给予高脂饮食喂养8周建立肥胖大鼠模型,采用随机数字表法从造模成功的大鼠中抽取24只,分成模型组、电针组、针加抑组和抑制剂组,每组各6只。造模结束后,电针组选取“中脘”“关元”“足三里”“丰隆”穴,其中“足三里”“丰隆”穴左右交替针刺。采用HANSLH202H型电针治疗仪,频率2 Hz,强度1 mA,连续波,留针10 min。针加抑组和抑制剂组大鼠给予SIRT1特异性抑制剂Nicotinamide,1 mg/kg,尾静脉注射给药,此外,针加抑组还另外实施与电针组相同的电针治疗。隔日1次,每周3次,共干预8周。分别在干预前和干预8周后测量大鼠体质量。各组大鼠处死前,心尖取血检测血清白介素6(IL-6)和肿瘤坏死因子α(TNF-α)的含量,采用Western blot法检测肝脏组织IL-6和TNF-α的蛋白表达。结果:与正常组相比,模型组大鼠体质量、肝脏组织中IL-6和TNF-α蛋白表达量均显著升高(P<0.01),血清IL-6和TNF-α含量均明显升高(P<0.05);与模型组相比,电针组、针加抑组大鼠体质量、肝脏组织中IL-6和TNF-α蛋白表达量在电针干预后显著降低(P<0.01),血清IL-6、TNF-α含量均明显下降(P<0.05);与抑制剂组相比,针加抑组大鼠体质量和肝脏组织中IL-6蛋白表达量降低(P<0.05,P<0.01),血清IL-6和TNF-α含量和TNF-α蛋白表达量均有下降趋势,但其差异无统计学意义(P>0.05)。结论:电针能够通过调控SIRT1改善大鼠肥胖状态,其机制可能与改善外周炎症水平有关。 Objective:To observe the effect of electro-acupuncture on liver inflammation in obese rats with insulin-resistant(IR)by regulating SIRT1 pathway,and to explore the mechanism of electro-acupuncture in improving obesity status in rats.Methods:70 male Wistar rats were adaptively fed for one week,10 rats were randomly selected to receive the standard diet,and 6 rats were randomly selected as the normal control after 8 weeks.The rest rats were fed with a high-fat diet for 8 weeks to establish the obese rat model,and 24 of the rats with the successful modeling were randomly selected and divided into the model group,the electro-acupuncture group,the electro-acupuncture combined with inhibitor group and the inhibitor group,with 6 rats in each group.The electro-acupuncture group was treated with electro-needling Zhongwan(CV12),Guanyuan(CV4),Zusanli(ST36)and Fenglong(ST40),among which ST36 and ST40 were alternately needled.SIRT1 specific inhibitor Nicotinamide was applied in the electro-acupuncture combined with inhibitor group and in the inhibitor group.Body mass was measured before and after 8 weeks of the intervention.Serum levels of interleukin-6(IL-6)and tumor necrosis factorα(TNF-α)were detected,and protein expressions of IL-6 and TNF-αin liver tissues were measured by Western blot method.Results:Compared to those in the normal group,the body mass and the expression levels of IL-6 and TNF-αin the liver tissues were significantly increased(P<0.01),and the serum levels of IL-6 and TNF-αwere significantly increased in the model group(P<0.05).Compared to those in the model group,the body mass,the expression levels of IL-6 and TNF-αin the liver tissues and the serum levels of IL-6 and TNF-αwere significantly decreased in the electro-acupuncture group and in the electro-acupuncture combined with inhibitor group after the treatment(P<0.01,P<0.05).Compared to those in the inhibitor group,the body mass and the protein expression level of IL-6 in the liver tissues were significantly decreased in the electro-acupuncture combined with inhibitor group(P<0.05,P<0.01);the serum contents of IL-6 and TNF-α,as well as TNF-αprotein were decreased in the electro-acupuncture combined with inhibitor group,but no statistical differences were seen between the two groups(P>0.05).Conclusion:Electro-acupuncture can regulate SIRT1 to improve the obesity status of the rats,which may be related to the improvement of peripheral inflammations.
