UPLC-MS/MS法快速测定龟甲胶中龟源成分

A Rapid UPLC-MS/MS Method for Identiflcation of Tortoise-derived Components in Testudinis Carapacis Et Plastri Colla

摘要 目的建立龟甲胶中龟源成分的快速检测方法。方法采用胰蛋白酶对龟甲胶、黄明胶、猪皮胶、阿胶进行酶解处理,利用超高效液相色谱一串联质谱检测技术对样品进行检测,通过对比质谱图进行龟源成分特征肽的筛选,利用龟源成分特征肽进行检测。结果找出龟甲胶的一条特征肽,其氨基酸序列为VGPTGPAGAR,其保留时间为4.77min,特征肽浓度0.1~1.2μg/mL范围内线性关系R^(2)为0.9998,检出限0.012μg/mL。结论所建方法专属性强、灵敏度高、线性良好,可快速实现对龟甲胶中龟源成分的检测分析,从而有效控制龟甲胶质量。 Objective To establish a rapid method for the determination of tortoise-Testudinis Carapacis et Plastri Colla.Methods After Testudinis carapacis et plastri colla,pig-skin gelatin,cattle-hide gelatin,and dongkey-hide gelatin were hydrolyzed by trypsin,ultra-high performance liquid chromatography-tandem mass spectrometry(UPLC-MS)was used to detect these samples.By comparing the mass spectras,a marker peptide of Testudinis Carapacis et Plastri Colla was screened out.Results A marker peptide of testudinis carapacis et plastri colla was identified.Its amino acid sequence was VGPTGPAGAR,and the retention time was 4.77 min.The linear relationship R^(2) was 0.9998 in the range of 0.1~1.2 μg/mL,and the detection limit was 0.012 μg/mL.Conclusion The method is specific,sensitive and linear.It can be used to quickly detect the tortoise-derived components in testudinis carapacis et plastri colla,and effectively control the quality of Testudinis Carapacis et Plastri Colla.
