摘要
目的:比较桑不同药用部位(桑叶、桑枝、桑白皮)总生物碱对α-葡萄糖苷酶活性的抑制作用。方法:采用采用蒸发光散射检测器(ELSD)-高效液相色谱法(HPLC)测定桑不同药用部位总生物碱中1-脱氧野尻霉素(DNJ)含量;以半数抑制浓度(IC 50)为评价指标,以阿卡波糖为阳性对照,以对-硝基苯基-α-D-吡喃葡萄糖苷为底物,采用体外抑制模型评价桑不同部位总生物碱对α-葡萄糖苷酶活性的抑制作用。结果:桑不同药用部位总生物碱中DNJ质量分数为桑白皮生物碱(30.1%)>桑枝生物碱(25.8%)>桑叶生物碱(21.4%)。桑不同药用部位总生物碱对α-葡萄糖苷酶抑制作用强度为桑枝生物碱>桑叶生物碱>桑白皮生物碱>阿卡波糖。结论:在桑枝、桑叶、桑白皮总生物碱中,以桑枝总生物碱对α-葡萄糖苷酶活性抑制作用最强,为桑资源开发辅助降血糖的药品和保健食品提供依据。
Objective:To compare inhibitory effects of alkaloids of different medicinal parts from Morus alba(leaf,root and twig)on the activities ofα-glycosidase.Methods:The content of 1-deoxynojirimycin(DNJ)in the total alkaloids in different medicinal parts from M.alba were determined by evaporative light scattering detector(ELSD)-high performance liquid chromatography(HPLC);Using half-inhibitory concentration value(IC 50)as evaluation index,acarbose as positive control,the inhibitory effects of alkaloids of different parts from M.alba on the activities ofα-glycosidase were evaluated with in vitro inhibition model.Results:The mass fraction of DNJ in total alkaloids in different medicinal parts from M.alba is Mori Cortex alkaloid(30.1%)>Mori Folium alkaloid(25.8%)>Mori Ramulus alkaloid(21.4%).In theα-glucosidase inhibitory activity test,the order of inhibitory activity was Mori Ramulus alkaloid>Mori Folium alkaloid>Mori Cortex alkaloid>acarbose.Conclusion:Among different parts of M.alba such as Mori Folium,Mori Cortex and Mori Ramulus,Mori Ramulus alkaloid shows the strongest inhibitory effects on the activities ofα-glucosidase,which has the value of being developed for the treatment of diabetes or health food.
