摘要
目的探讨ADRB1基因多态性与冠心病发病及病情轻重的关系。方法选取我院2017年3月—2019年5月收治的86例冠心病病人为试验组,选择同期健康体检者86名为对照组。采用聚合酶链反应结合DNA直接测序法检测ADRB1基因型,比较各组基因型和等位基因分布频率的差异。结果多因素Logistic回归分析结果表明,ADRB1基因Ser49Gly位点等位基因模型(Ser与Gly)和显性基因模型(Ser/Ser与Ser/Gly+Gly/Gly)为冠心病的影响因素[OR=2.4,95%CI(1.3,5.1),P=0.008;OR=1.9,95%CI(1.1,3.4),P=0.012];Arg389Gly位点各基因模型与冠心病无相关性;试验组SYNTAX评分3个亚组间Ser49Gly位点多态性分布比较差异均无统计学意义(P>0.05)。结论ADRB1基因Ser49Gly位点多态性与冠心病相关而与SYNTAX评分所示的冠心病病情轻重无关;基因Arg389Gly位点多态性与冠心病无独立相关。
Objective To explore the correlation between ADRB1 gene polymorphism and the incidence and severity of coronary heart disease.Methods Eighty-six patients with coronary heart disease were selected as the experimental group,and 86 healthy persons who underwent physical examination were selected as the control group at the same time.The genotypes of ADRB1 were detected by polymerase chain reaction(PCR)combined with direct DNA sequencing,and the distribution frequencies of genotypes and alleles in each group were compared.Results The results of multivariate Logistic regression analysis showed that the Ser49Gly allele model(Ser vs Gly)and the dominant gene model(Ser/Ser vs Ser/Gly+Gly/Gly)of ADRB1 gene were correlated with coronary heart disease[OR=2.4,95%CI(1.3,5.1),P=0.008;OR=1.9,95%CI(1.1,3.4),P=0.012].There was no correlation between the Arg389Gly gene model and coronary heart disease.There was no significant difference between the polymorphism distribution of Ser49Gly sites among 3 subgroups in the experimental group(P>0.05).Conclusion Ser49Gly sites of ADRB1 gene polymorphism was correlated with coronary heart disease,and without relation of the severity of coronary heart disease as shown in SYNTAX score.There was no independent correlation between Arg389Gly polymorphism and coronary heart disease.
