摘要 目的探讨RNA干扰GMF-β基因对脑梗死小鼠海马组织神经元凋亡及炎症水平的影响.方法选择SPF级小鼠40只,将小鼠随机分为假手术组、模型组、control-shRNA组、GMF-βshRNA组,每组10只.建模后进行小鼠神经功能缺损评分;qRT-PCR检测小鼠脑组织GMF-β基因的mRNA表达水平;TTC染色法测定小鼠脑梗死体积;TUNEL法测定小鼠脑组织神经元凋亡;ELISA法检测小鼠脑组织及血清中TNF-c、IL-6表达水平.结果与假手术组相比,模型组和control-shRNA组小鼠神经损伤明显,神经行为学评分显著升高,差异有统计学意义(P<0.05);与模型组和control-shRNA组相比,GMF-βshRNA组小鼠神经损伤明显改善,神经行为学评分显著降低(P<0.05).与假手术组相比,模型组小鼠脑梗死体积、脑组织神经元凋亡细胞数、脑组织及血清中TNF-α、IL-6表达水平均显著升高(均P<0.05);抑制GMF-β的表达后,小鼠神经行为学评分、脑梗死体积、脑组织神经元凋亡细胞数、脑组织及血清中TNF-α、IL-6表达水平均显著减少(P<0.05),神经损伤明显改善.结论抑制GMF-β的表达可通过降低脑梗死小鼠脑海马组织梗死体积、神经元凋亡水平及机体炎症水平,从而改善脑梗死小鼠脑组织损伤. Objective To investigate the effect of RNA interference with GMF-βgene on neuron apoptosis and inflammation in hippocampus of mice with cerebral infarction.Methods Forty SPF mice were randomly divided into sham operated group,model group,control shRNA group and GMF-βshRNA group.Neurological deficit score of mice after modeling.qRT-PCR detection of GMF-βgene mRNA expression in mouse brain.Determination of cerebral infarction volume in mice by TTC staining.TUNEL method for the determination of neuron apoptosis in mouse brain.Detection of TNF-a and IL-6 expression in brain and serum of mice by ELISA.Results Compared with the sham operation group,the model group and the control shRNA group had significant neurological injury and neurobehavioral scores,the difference was statistically significant(P<0.05);compared with the model group and the control shRNA group,the GMF-βshRNA group had significant improvement in neurological injury and significantly reduced neurobehavioral scores(P<0.05).Compared with sham operation group,the volume of cerebral infarction,number of apoptotic neurons in brain tissue,TNF-a and IL-6 expression level in brain tissue and serum in model group were significantly increased(P<0.05);after inhibiting the expression of GMF-β,neurobehavioral score,cerebral infarction volume,number of apoptotic neurons in brain tissue,TNF-a and IL-6 expression level in brain tissue and serum were significantly decreased(all P<0.05).The nerve injury was significantly improved.Conclusion Inhibition of GMF-βexpression can improve the brain damage of cerebral infarction mice by reducing the infarct volume,neuron apoptosis and body inflammation.
