摘要 目的:通过体外培养人乳腺癌难愈合创面中成纤维细胞,并应用TGF-β/Smad信号通路阻断剂干扰素γ(IFN-γ)干扰后,探讨IFN-γ在乳腺癌难愈合创面中所发挥的作用和影响。方法:采用酶消化法对人乳腺癌溃疡创面组织行体外成纤维细胞原代培养。将实验细胞分成A、B、C、D共四组,A组为IFN-γ5000U/ml组;B组为IFN-γ10000U/ml组;C组为对照组,不加药物;D组为空白组。分别于第7天、第14天将培养细胞接种于96孔板内,用MTT法测定细胞增殖情况。结果:第7天及第14天A组吸光值与对照组C组比较差异无统计学意义(P>0.05);B组吸光值均低于对照组C组,差异具有统计学意义(P<0.05)。结论:IFN-γ对成纤维细胞的增殖抑制受到浓度的影响,浓度10000U/ml时明显抑制了体外培养成纤维细胞的增殖,而在较低浓度(5000U/ml)时对体外培养成纤维细胞增殖无明显抑制作用。 Objective The role and influence of IFN-γin refractory wounds of breast cancer were investigated by in vitro culture of fibroblast cells in refractory wounds of breast cancer and interference with IFN-γ(TGF-β/Smad signaling pathway blocker).Methods In vitro fibroblast primary culture was performed on the wound tissue of human breast cancer ulcer by enzyme digestion.The experimental cells were divided into four groups:group A,group B,group C and group D.Group A was IFN-γ5000U/ml group,group B was IFN-γ10000U/ml group;group C was the control group without drugs,group D was blank group.The cultured cells were inoculated into 96-well plates on the 7th and 14th day respectively,and cell proliferation was determined by MTT method.Results There was no significant difference in OD value between group A and control group C on the 7th and 14th day(P>0.05).The OD value of group B was lower than those of group C,the differences were statistically significant(P<0.05).Conclusion The inhibition of IFN-γon proliferation of fibroblasts was affected by the concentration,the concentration at 10000U/ml significantly inhibited proliferation of fibroblasts cultured in vitro,while the lower concentration of IFN-γat 5000U/ml no obvious inhibitory effect on the proliferation of fibroblasts.
