摘要
目的探讨人脐带来源的间充质干细胞(HUC-MSCs)对小鼠溃疡性结肠炎(UC)的作用。方法小鼠自由饮用3.5%葡聚糖硫酸钠水溶液构建UC模型。按照体重将小鼠随机分为6组:正常组、模型组、对照组和低、中、高3个剂量实验组,每组10只。正常组与模型组腹腔注射高糖DMEM培养液;对照组给予美沙拉嗪100 mg·kg^(-1)灌胃;低、中、高3个剂量实验组按照每只1×10^(6),5×10^(6)和1×10^(7)腹腔注射HUC-MSCs, 1次/天,连续6 d。收集并评估小鼠体重和结肠长度,用酶联免疫吸附法检测结肠组织γ-干扰素(IFN-γ)、肿瘤坏死因子-α(TNF-α)和白细胞介素-10(IL-10)的含量。结果正常组、模型组和低、中、高3个剂量实验组的IFN-γ含量分别为(5.97±0.92),(12.05±0.63),(5.93±0.93),(9.52±0.54),(9.16±0.80)和(8.58±0.60)ng·mg^(-1);这5组的TNF-α含量分别为(3 764.80±568.59),(7 803.80±343.11),(3 535.70±429.73),(6 257.70±528.17),(5 926.60±430.47)和(5 284.00±481.41)pg·mg^(-1);这5组的IL-10含量分别为(466.04±37.11),(136.51±18.36),(423.56±30.58),(257.35±26.21),(288.14±24.31)和(329.95±27.73)pg·mg^(-1)。上述这3个指标:模型组与正常组比较,差异均有统计学意义(均P<0.01);不同剂量实验组与模型组比较,差异均有统计学意义(均P<0.01)。结论 HUC-MSCs可明显改善UC病变,减轻炎症反应。
Objective To investigate the effects of human umbilical cord-derived mesenchymal stem cells(HUC-MSCs)on ulcerative colitis(UC)in mice.Methods UC model was established by adding 3.5%dextran sodium sulfateto drinking water for mice.The mice were randomly divided into 6 groups:normal group,model group,control group,as well as low,medium and high dose of experimental groups,with 10 mice in each group.Mice in normal group and model group were intraperitoneally injected with high glucose DMEM culture medium,mice in control group were orally treated with mesalazine(100 mg·kg^(-1)),while mice in experimental groups were injected intraperitoneally with HUC-MSCs at the dose of 1×106 cells,5×106 cells and 1×107 cells,respectively.All the drugs were administered for consecutive 6 d,once a day.Body weight and colon length were assessed on day 7.The contents of interferonγ(IFN-γ),tumor necrosis factorα(TNF-α)and interleukin-10(IL^(-1)0)in the colon were determined by enzyme linked immunosorbent assay.Results The INF-γcontent of normal group,model group,and the three dose of experimental groups were(5.97±0.92),(12.05±0.63),(5.93±0.93),(9.52±0.54),(9.16±0.80)and(8.58±0.60)ng·mg^(-1) respectively;the TNF-αcontent in the 5 groups were(3764.80±568.59),(7803.80±343.11),(3535.70±429.73),(6257.70±528.17),(5926.60±430.47)and(5284.00±481.41)pg·mg^(-1),respectively;the IL^(-1)0 content in the 5 groups were(466.04±37.11),(136.51±18.36),(423.56±30.58),(257.35±26.21),(288.14±24.31)and(329.95±27.73)pg·mg^(-1),respectively.Significant difference of the factors were found between normal group and model group(all P<0.01),and significant difference of the factors were also found between the experimental groups and model group(all P<0.01).Conclusion HUC-MSCs can significantly improve UC lesions and reduce inflammation.
