摘要
目的检测妊娠早期绒毛及蜕膜组织中Storkhead box1(STOX1)mRNA和蛋白表达。方法绒毛和蜕膜组织来自60例不同孕龄的健康孕妇(6~11孕周),按孕周分为6组,每组10例,分别采用免疫组织化学法、逆转录聚合酶链反应法和Western blotting法检测STOX1的定位、mRNA及蛋白表达水平。结果在妊娠早期各孕周绒毛和蜕膜组织中均可检测到STOX1的表达。在绒毛滋养层,mRNA表达在孕6周时最低,随孕周增加表达逐渐上升,孕11周最高,各相邻孕周间差异无统计学意义(P>0.05),但间隔孕周之间差异有统计学意义(P<0.05),蛋白表达与mRNA表达趋势相似。蜕膜组织中,mRNA与蛋白均稳定表达,各孕周间无明显差异。结论STOX1在妊娠早期绒毛和蜕膜层表达不同,提示其可能参与了正常妊娠早期滋养层的发育和子宫内膜蜕膜化的调控。
Objective To study the mRNA and protein expression of Storkhead box1(STOX1)in normal villi and deciduas tissues during early trimester of pregnancy.Methods Villi and deciduas tissues were obtained from 60 cases healthy pregnant women,who were divided into 6 groups(10 cases in each group)according to the gestational weeks(6~11 weeks),the localization and expression of STOX1 mRNA and protein levels were detected by immunohhistochemistry,early trimester of pregnancy and Western blotting.Results STOX1 was positively expressed in the villi and deciduas tissues of all groups during early trimester of pregnancy.In villi tissues,the expression of STOX1 mRNA and protein was the lowest in 6 weeks of gestation and increased with the progress of gestation weeks,the highest was in 11 weeks of gestation.There was no statistical difference of STOX1 level between adjacent groups(P>0.05),while there was significant difference of STOX1 level between the interval groups(P<0.05).In decidua tissues,there was no significant difference of STOX1 mRNA and protein in different gestational weeks.Conclusion The different expression,of STOX1 in villi and decidua tissues suggestted that STOX1 gene may play an important role in regulating the development of trophoblast and decidualization of endometrial stromal cells during early trimester of pregnancy.
