摘要
目的:探讨赖氨酸特异性脱甲基酶2B(KDM2B)与p21在卵巢癌组织中的表达及其临床意义。方法:采用回顾性研究方法,收集20例正常卵巢组织、33例良性卵巢组织和59例恶性卵巢组织。采用免疫组化和实时荧光定量PCR(qPCR)方法分别检测3种不同卵巢组织KDM2B和p21 mRNA及蛋白的表达情况,并分析两者在卵巢癌中的表达相关性及两者与卵巢癌患者临床病理特征的关系。结果:恶性卵巢组织中KDM2B mRNA及蛋白表达阳性率显著高于良性卵巢组织、正常卵巢组织(P<0.05),而p21 mRNA及蛋白表达阳性率显著低于良性卵巢组织、正常卵巢组织(P<0.05)。p21在低分化、FIGO分期Ⅲ~Ⅳ期及有淋巴结转移的卵巢癌组织中的蛋白表达阳性率低于中、高分化、FIGO分期Ⅰ~Ⅱ期、无淋巴结转移(P<0.05),而KDM2B蛋白表达情况相反(P<0.05)。在恶性卵巢组织中,KDM2B与p21蛋白表达呈负相关关系(r=-0.978,P=0.000)。结论:KDM2B在恶性卵巢组织中高表达,p21在恶性卵巢组织中低表达,两者均与肿瘤组织分级、FIGO分期及淋巴结转移有关。
Objective:To investigate the expression and clinical significance of lysine-specific demethylase 2B(KDM2B)and p21 in ovarian cancer.Methods:A total of 20 normal ovarian tissues,33 benign ovarian tissues and 59 malignant ovarian tissues were collected retrospectively.The mRNA and protein expressions of KDM2B and p21 in three ovarian tissues were detected by Immunohistochemistry(IHC)andqPCR,respectively.The correlation between the two expressions in ovarian cancer and the clinicopathological characteristics of ovarian cancer patients were analyzed.Results:The protein and mRNA expression of KDM2B in malignant ovarian tissues were higher than that in normal ovarian tissues and begin ovarian tissues,wihle the protein and mRNA expression levels of p21 was lower(P<0.05).The positive rate of p21 protein expressionin poorly differentiated,FIGO stageⅢ-Ⅳand lymph node metastasis cases were lower than those in high-medium differentiated,FIGO stageⅠ-Ⅱand no lymph node metastasis cases(P<0.05).While the expression of KDM2B protein was the opposite(P<0.05).In malignant ovarian tissues,KDM2B was negatively correlated with the protein expression of p21(P<0.05).Conclusions:KDM2B was highly expressed in malignant ovarian tissues,while p21 was low expressed in malignant ovarian tissues,both of which were related to tumor grade,FIGO stage and lymph node metastasis.
