miR-362-3p和MCAM在稽留流产患者绒毛组织中的表达及其与微血管密度的关系 被引量:2

摘要 目的:探讨miR-362-3p和人黑色素瘤细胞黏附分子(MCAM)在稽留流产患者绒毛组织中的表达及其与微血管密度(MVD)的关系。方法:选取30例稽留流产患者作为稽留流产组,人工流产的30例早孕孕妇作为对照组。运用生物信息学预测到miR-362-3p的一个靶基因MCAM,采用实时荧光定量PCR(qPCR)检测两组绒毛组织中miR-362-3p和MCAM mRNA的表达水平,随机选取两组各20例采用免疫组织化学法(IHC)检测绒毛组织中MCAM的表达和MVD。结果:miR-362-3p在稽留流产组中的相对表达水平高于人工流产组(P<0.05);MCAM mRNA在稽留流产组中的相对表达水平低于人工流产组(P<0.05);稽留流产组中miR-362-3p和MCAM mRNA的相对表达量呈负相关关系(r=-0.796,P<0.05)。MVD在稽留流产组中的表达水平低于人工流产组(P<0.05),MCAM的蛋白表达量与MVD呈正相关关系(r=0.667,P<0.05)。结论:miR-362-3p可能通过调控MCAM影响胎盘MVD,从而与胎盘发育过程中稽留流产的发生发展相关。 Objective:To investigate the expression of miR-362-3p and human melanoma cell adhesion molecules(MCAM)in the villi of patients with missed abortion and their relationship with microvascular density(MVD).Methods:A total of30 cases of missed abortion patients were selected as the missed abortion group,and 30 cases of induced abortion patients in early pregnancy were collected as the control group.MCAM,a target gene of miR-362-3p,was predicted using bioinformatics.Real-time quantitative reverse transcription polymerase chain reaction(qPCR)was used to detect the expression levels of miR-362-3p and MCAM mRNA in the villi of the two groups.Immunohistochemical(IHC)was used to randomly select 20 cases in each group to detect MCAM expression and microvascular density in the villi of the two groups.Results:Therelative expression of miR-362-3p in missed abortion tissues was higher than that in induced abortion tissues(P<0.05);MCAM mRNA relative expression in missed abortion tissues was lower than that in induced abortion tissues(P<0.05).MVD in missed abortion tissue samples was significantly lower than that in induced abortion(P<0.05).The relative expression levels of miR-362-3p and MCAM mRNA in the aborted villi were negatively correlated(r=-0.796,P<0.05),The protein expression of MCAM was positively correlated with MVD(r=0.667,P<0.05).Conclusion:MiR-362-3p may affect the microvessel density by regulating MCAM,which is related to the occurrence and development of missed abortion during placental development.
