当归药材及当归身饮片高效液相色谱特征图谱研究 被引量:4

摘要 目的:建立当归药材及当归身饮片高效液相色谱(HPLC)特征图谱,并通过对当归药材和当归身饮片特征图谱的比较分析,为当归药材和当归身饮片的质量控制提供参考依据。方法:采用Symmetry C_(18)色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.2%乙酸为流动相,梯度洗脱,检测波长为280 nm,柱温为30℃,流速为1.0 mL/min,进样量20μL;采用“中药色谱指纹图谱相似度评价系统”对3产地15批当归药材及相应当归身饮片进行相似度评价,建立当归药材及当归身饮片HPLC特征图谱。结果:以阿魏酸为参照峰,构建了由阿魏酸、洋川芎内酯I、洋川芎内酯H和藁本内酯4个特征峰组成的当归药材及当归身饮片HPLC特征图谱,15批当归药材相似度>0.96,15批对应当归身饮片相似度>0.97,当归药材与当归身饮片特征图谱相似,但各成分含量高低存在差异。结论:本研究建立的方法稳定可靠,重现性好,可为当归药材和当归身饮片质量控制提供参考,为含当归或当归身的经典名方的开发研究提供依据。 Objective:To establish the HPLC specific chromatogram of Angelica sinensis and Angelica sinensis body,and to provide reference for the quality control of Angelica sinensis and Angelica body by the comparative analysis of the characteristic maps of Angelica sinensis and Angelica body.Methods:The HPLC analysis was performed on an Symmetry C_(18) column(250 mm×4.6 mm,5μm)with acetonitrile-0.2%acetic acid solution as the mobile phase with gradient elution at a flow rate of 1.0 mL/min.The detection wavelength was 280 nm.The column temperature was 30℃.The flow rate was 1.0 mL/min,and the injection volume was 20μL;Similarity evaluation system of chromatographic fingerprint of Chinese medicine was used to evaluate the similarity of 15 batches of Angelica sinensis and Angelica body in 3 production areas,and to establish the characterisics fingerprint of Angelica sinensis and Angelica body.Results:The specific chromatogram was preliminarily constructed and 4 common peaks of ferulic acid,senkyunolide I,senkyunolide H and ligustilide with ferulic acid as the reference were identified.The similarity of 15 batches of Angelica sinensis was greater than 0.96,and the similarity of 15 batches of angelica body was greater than 0.97.The characteristics of the HPLC specific chromatogram of Angelica sinensis and angelica body were similar,but the content of each component was different.Conclusion:The method is stable,reliable and reproducible,and can be used for quality control of Angelica sinensis and angelica body,and provide a basis for the development of famous classical formulas containing Angelica sinensis or angelica body.
