鼠神经生长因子联合丹参川芎嗪注射液对缺氧缺血性脑病新生儿氧化应激及炎症因子水平的影响 被引量:2

摘要 目的探讨鼠神经生长因子联合丹参川芎嗪注射液对缺氧缺血性脑病新生儿氧化应激及炎症因子水平的影响。方法将98例缺氧缺血性脑病新生儿按随机数字表法分为两组,每组49例。两组新生儿均给予常规对症治疗,在此基础上,对照组给予鼠神经生长因子治疗,观察组给予鼠神经生长因子联合丹参川芎嗪注射液治疗。观察2周。比较两组临床疗效及不良反应发生率,统计治疗前后两组新生儿意识障碍、原始反射和肌张力恢复时间。治疗前后,应用酶联免疫吸附法检测血清白细胞介素-1β、白细胞介素-6、白细胞介素-18水平,应用高通量逆转录实时荧光定量测定Nod样受体蛋白3、凋亡相关斑点样蛋白、半胱氨酸天冬氨酸特异蛋白酶mRNA水平。结果观察组临床疗效显著优于对照组(P<0.05)。观察组新生儿意识障碍、原始反射和肌张力恢复时间显著短于对照组(P<0.01),白细胞介素-1β、白细胞介素-6、白细胞介素-18水平以及Nod样受体蛋白3、凋亡相关斑点样蛋白、半胱氨酸天冬氨酸特异蛋白酶mRNA水平显著低于对照组(P<0.01)。两组不良反应发生率比较差异无统计学意义(P>0.05)。结论鼠神经生长因子联合丹参川芎嗪注射液治疗新生儿缺氧缺血性脑病临床疗效明显,能更好地调节患儿白细胞介素-1β、白细胞介素-6、白细胞介素-18以及Nod样受体蛋白3、凋亡相关斑点样蛋白、半胱氨酸天冬氨酸特异蛋白酶mRNA水平。 Objective To observe the effect of rat nerve growth factor combined with Danshen ligustrazine injection on oxidative stress and inflammatory factors in neonates with hypoxic-ischemic encephalopathy.Methods A total of 98 newborns with hypoxic-ischemic encephalopathy were divided into two groups,each with 49 cases.Both groups of newborns were given conventional symptomatic treatment.On this basis,the control group was given rat nerve growth factor treatment,and the observation group was given rat nerve growth factor combined with Danshen ligustrazine injection for 2 weeks.The clinical efficacy and the incidence of adverse reactions were compared between the two groups.The recovery time of consciousness disturbance,primitive reflex and muscle tone of the two groups of newborns before and after treatment was counted.Before and after treatment,the enzyme linked immunosorbent assay(ELISA)was used to detect serum levels of interleukin-1β(IL-1β),interleukin-6(IL-6),and interleukin-18(IL-18).The real-time quantitative reverse transcription polymerase chain reaction(QRT-PCR)was used to determine the NOD-like receptor protein3(NLRP3),apoptosis associated speck like protein containing a CRAD(ASC),cysteinyl aspartate-specific proteinase-1(Caspase-1)mRNA levels.Results The clinical efficacy of the observation group was significantly better than that of the control group(P<0.05).The recovery time of consciousness disturbance,primitive reflex and muscle tone in the observation group was significantly shorter than that in the control group(P<0.01),the levels of IL-1β,IL-6,IL-18 and NLRP3,ASC,Caspase-1 mRNA levels were significantly lower than those of the control group(P<0.01).There was no significant difference in the incidence of adverse reactions between the two groups(P>0.05).Conclusions Rat nerve growth factor combined with Danshen ligustrazine injection has obvious clinical effects in the treatment of neonatal hypoxic-ischemic encephalopathy,and can better regulate IL-1β,IL-6,IL-18 and NLRP3,ASC,Caspase-1 mRNA levels in children.
