摘要
本文提出了一种基于超声辅助悬浮有机液滴固化微萃取结合紫外分光光度法(UV)测定药片及血清中ACC007含量的方法。在超声辅助下,萃取剂1-十一醇在待测样品的水溶液中产生乳化现象,并能有效快速地萃取水相中的ACC007。将萃取液离心后冰水浴中冷却,1-十一醇凝固漂浮在水面可方便地与溶液分离,最后用乙醇溶解1-十一醇后用UV法测定萃取剂中ACC007的含量。实验中对影响萃取率的几个重要参数如溶液pH、萃取剂的用量、超声时间、离心时间、冷却时间等进行了优化。在优化条件下,方法的线性范围为0.50-20.0μg·mL^(-1),线性方程为A=0.0486 c+0.0421(c:μg·mL^(-1)),相关系数R=0.998,检出限为0.087μg·mL^(-1),加标回收率为92.0%~104.3%。研究结果表明该方法可用于药片和人血清中ACC007含量的测定。
In this paper,a new method of ultrasound-assisted solidification of floating organic drop micro extraction coupled to UV spectrometry for the determination of ACC007 in tablet and serum was proposed.Under the assistance of ultrasound,the extractant 1-undecanol produced emulsification in sample solution and could extract ACC007 in the aqueous phase quickly and effectively.After centrifugation,the solution was cooled at ice bath and 1-undecanol was solidified and floating on the water,which made 1-undecanol easy to separate from aqueous solution.Finally the ACC007 in 1-undecanol extraction liquid was dissolved in ethanol and measured by UV spectrophotometry.Several important parameters affecting the extraction efficiency such as sample pH,the volume of extraction solvent,ultrasonic time,centrifugation time and cooling time were studied and optimized.Under the optimized conditions,there was good linear relationships for ACC007 over the concentration range of 0.50~20.0μg·mL^(-1).The linear equation was A=0.0486 c+0.0421(c:μg·mL^(-1))with correlation coefficient R=0.998 and the limit of detection was 0.087μg·mL^(-1),and the average recoveries were between 92.0%and 104.3%.The results showed the studied method could be used to determine ACC007 in tablet and human serum.
