胃癌预后预测生物标志物和治疗靶点的筛选与研究

Identification and Analysis of Prognostic Biomarkers and Therapeutic Targets in Human Gastric Cancer

目的:挖掘有前景的生物标志物准确地预测胃癌患者的总生存,寻找新的胃癌治疗靶点改善患者预后。方法:通过NCBI-GEO数据库中包含胃癌组织和正常组织基因表达谱的芯片,筛选出胃癌组织中相比正常胃组织高表达的差异基因。再借助GEPIA数据库验证这基因的表达。通过Kaplan-Meier plotter数据库分析基因表达的高低与患者预后的相关性。最后借助HCMBD数据库分析基因的功能。结果:筛选出26个在胃癌组织中高表达的基因,进一步通过GEPIA数据库证明其中25个基因在胃癌中高表达,还有22个基因的表达随着胃癌疾病进展表达逐渐升高。这25个高表达的基因在胃癌患者中进行生存分析显示有23个基因的表达与胃癌患者总生存期显著负相关。结合这些基因的具体功能,最终选出7个基因:BGN、CTHRC1、INHBA、THBS2、THY1、TIMP1、WISP1,有希望成为胃癌预后预测生物标志物和潜在的治疗靶点。结论:我们筛选出了与胃癌发展及预后高度相关的关键基因,有助于进一步了解胃癌的分子基础,指导临床胃癌患者复查频率和开发新的靶向治疗。

Objective:To discover promising biomarkers to more accurately predict the survival of patients with GC,and to provide novel insights to design new targeted agents for GC treatment.Methods:Gene expression information of GC from GEO database and obtained significantly highly expressed genes in gastric tumor tissues were compared with normal controls.The mRNA expression of these genes in GC tissues and normal controls were detected via GEPIA databases.The prognostic value of these highly expressed genes was analyzed using Kaplan-Meier plotter database.The function of these highly expressed genes were analyzedvia HCMBD database.Results:There were 26 significantly highly expressed genes in gastric tumor tissues compared with normal controls.All these genes except FDLIM7 were highly expressed in GC tissues analyzed via GEPIA databases.In addition,significant correlations were found between the expression of 22 genes and the pathological stage of patients with GC.The expressions of 23 genes were associated with a significantly worse prognosis.Based on the function of these 26 genes.BGN,CTHRC1,INHBA,THBS2,THY1,TIMP1 and WISP1 mightbe promising prognostic biomarkers and potential therapeutic targets for GC.Conclusion:Our study screened out key genes highly associating with GC.s developments and prognosis,and it is useful to further understand the molecular basis of gastric cancer and guide the frequency of postoperative re-examination and treatment for patients.
