黄芪颗粒对老年痴呆大鼠海马区血脑屏障转运相关蛋白的影响 被引量:2

Effect of Astragalus Granule on blood brain barrier transport related proteins in hippocampus of Alzheimer’s disease rats

摘要 目的研究P-糖蛋白(P-gp)和低密度脂蛋白受体相关蛋白(LRP-1)在老年痴呆(AD)大鼠海马组织中的含量,经过口服黄芪颗粒后分析两者含量的变化来探讨黄芪颗粒的作用机制。方法建造AD大鼠模型,将AD大鼠分为AD组、黄芪颗粒组和西药组,同时选正常的同数量的大鼠作为对照组。黄芪颗粒组和西药组分别口服黄芪颗粒和吡拉西坦,对照组和AD组口服同容量三蒸水,口服时长为30天。口服用药前、后分别检测4组大鼠的行为能力的改变情况,Western blot测量口服用药后4组大鼠海马组织中P-gp和LRP-1的含量。结果口服用药前,AD组、黄芪颗粒组和西药组大鼠的行为能力均弱于对照组;口服用药后,AD组大鼠的行为能力弱于对照组(均P<0.05),海马组织中P-gp(0.045±0.004)和LRP-1(0.167±0.012)的含量少于对照组(均P<0.05);黄芪颗粒组大鼠的行为能力强于AD组(均P<0.05),海马组织中P-gp(0.092±0.007)和LRP-1(0.235±0.014)的含量多于AD组(均P<0.05);黄芪颗粒组与西药组相比,差异无统计学意义(P>0.05)。结论口服黄芪颗粒后能增加AD大鼠海马组织中P-gp和LRP-1的含量来达到改善AD大鼠的行为能力。 Objective To study the content of P-glycoprotein(P-gp) and low-density lipoprotein receptor related protein-1(LRP-1)in the hippocampus of Alzheimer’s disease(AD) rats, explore the mechanism of Astragalus Granule by analyzing the changes of the two contents after oral administration. Methods AD rat model was established, and the AD rats were arranged into AD group,Astragalus Granules group and western medicine group, at the same time, the same number of normal rats were selected as the control group. Astragalus Granules group and western medicine group treated with Astragalus Granules and piracetam by oral administration respectively, while the control group and AD were treated with the same volume of three steaming water orally,and the treatment was for 30 days. Before and after oral administration, the behavior ability of the four groups were detected.Western blot was used to measure the contents of P-gp and LRP-1 in hippocampus of rats in four groups after oral administration.Results Before oral administration, the behavior ability of AD group, Astragalus Granules group and western medicine group was weaker than that of the control group. After oral administration, the behavior ability of AD group was weaker than that of control group(P<0.05), and the content of P-gp(0.045±0.004) and LRP-1(0.167±0.012) in hippocampus was lower than that of control group(P<0.05). The behavior ability of rats in Astragalus Granules group was stronger than that in AD group(all P<0.05), the content of P-gp(0.092±0.007) and P-gp(0.235±0.014) in hippocampus was higher than that in AD group(all P<0.05). The difference was not statistically significant between Astragalus Granules group and western medicine group(P>0.05). Conclusion The oral administration of Astragalus Granules can raise the content of P-gp and LRP-1 in the hippocampus of AD rats, so as to improve the behavior of AD rats.
