枸杞多糖对慢性睡眠剥夺大鼠海马区去甲肾上腺素及其受体含量的影响 被引量:4

Effects of Lycium barbarum polysaccharide on contents of norepinephrine and norepinephrine receptor in hippocampus of rats with chronic sleep deprivation

摘要 目的分析枸杞多糖(LBP)对慢性睡眠剥夺大鼠行为能力和海马组织中去甲肾上腺素及其受体含量的影响。方法实验安排为4组,正常组、睡眠剥夺组、LBP干预组及脑复康组。正常组:常规饮食;睡眠剥夺组:对大鼠实行每天剥夺18 h的睡眠,连续进行30 d;LBP干预组和脑复康组:在大鼠进行睡眠剥夺的同时口服LBP 5 mL/100 mg·kg。评估各组大鼠的行为能力指标的变化,检测海马组织中去甲肾上腺素的含量,免疫印迹法衡量海马组织中α1和β肾上腺素能受体(α1-AR和β-AR)的含量。结果与正常组相比,睡眠剥夺组行为能力减弱,海马组织中去甲肾上腺素及α1-AR(0.139±0.024)和β-AR(0.141±0.024)的含量下降(均P<0.05);与睡眠剥夺组相比,LBP干预组的行为能力增强,海马组织中去甲肾上腺素及α1-AR(0.211±0.017)和β-AR(0.215±0.015)的含量升高(均P<0.05);LBP干预组与脑复康组之间差异无统计学意义(P>0.05)。结论 LBP能改善慢性睡眠剥夺大鼠的行为能力,这与其能提高慢性睡眠剥夺大鼠海马组织中去甲肾上腺素及其α1-AR和β-AR的含量密切相关。 Objective To analyze the effects of Lycium barbarum polysaccharide(LBP) on the behavioral ability, norepinephrine and norepinephrine receptor in hippocampus of rats with chronic sleep deprivation. Methods The experiment was arranged into four groups: normal group, sleep deprivation group, LBP intervention group and Naofukang group. Normal group: rats were given normal diet. Sleep deprivation group: rats were deprived of sleep for 18 hours a day for 30 consecutive days. LBP intervention group and Naofukang group: rats were deprived of sleep at the same time of oral LBP 5 mL/100 mg·kg. The changes of behavior ability of rats in each group were evaluated. The norepinephrine content in hippocampus was detected, and the contents of α1 and β adrenergic receptor(α1-AR and β-AR) in hippocampus were measured. Results Compared with the normal group, the behavior ability of sleep deprived group decreased, and the norepinephrine content and the contents of α1-AR(0.139±0.024) and β-AR(0.141±0.024) in hippocampus decreased(all P<0.05). Compared with the sleep deprivation group, the behavioral ability of LBP group was enhanced, and the norepinephrine content and the contents of α1-AR(0.211±0.017) and β-AR(0.215±0.015) in hippocampus increased(all P<0.05). There was statistically significant difference between LBP group and Naofukang group(P>0.05). Conclusion LBP can improve the behavior ability of rats with chronic sleep deprivation, which is closely related to it increases the norepinephrine content and the contents of α1-AR and β-AR in hippocampus of sleep deprivation rats.
