摘要
目的从抑制神经炎症探讨酸枣仁汤减轻睡眠剥夺大鼠海马神经损伤的机制。方法将大鼠随机分为正常对照组、模型对照组、艾司唑仑组、酸枣仁汤低剂量组、酸枣仁汤高剂量组。除正常对照组外,其他组构建睡眠剥夺动物模型,造模与给药同时进行,连续给药30d。应用实时荧光定量聚合酶链式反应(Real-time PCR)法与ELISA检测大鼠海马中白细胞介素-1β(IL-1β)、肿瘤坏死因子-α(TNF-α)mRNA与蛋白表达水平;尼氏染色法观察海马神经的病理形态。结果与正常组比较,模型组大鼠海马中IL-1βmRNA、TNF-αmRNA与蛋白表达水平显著升高(P<0.01,P<0.05);与模型组比较,酸枣仁汤低剂量组和酸枣仁汤高剂量组大鼠海马中IL-1βmRNA、TNF-αmRNA与蛋白表达水平均有不同程度降低(P<0.01,P<0.05),而艾司唑仑组大鼠海马中IL-1βmRNA、TNF-αmRNA与蛋白表达水平变化不显著(P>0.05)。模型组大鼠海马CA3区神经元排列欠规则,细胞周围间隙增宽胞质内尼氏小体明显减少酸枣仁汤低剂量组、酸枣仁汤高剂量组大鼠海马CA3区神经元排列较为规则,细胞周围间隙缩短尼氏体较为丰富;艾司唑仑组大鼠海马CA3区神经元排列紊乱和尼氏体数量减少改善不明显。结论酸枣仁汤可抑制神经炎症减轻睡眠剥夺大鼠海马神经损伤。
Objective To explore the mechanism of Suanzaoren Decoction in alleviating hippocampal nerve injury in sleep-deprived rats based on neuroinflammation.Methods The rats were randomly divided into normal control group,model group,eszolam group Suanzaoren Decoction low dose groupand Suanzaoren Decoction high dose group.In addition of normal control group,other groups constructed sleep deprivation model,which was conducted simultaneously with the drug administration,and the drug was continuously administered for 30 d.The expression levels of interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α)mRNA and protein in hippocampus of rats were detected by real-time PCR and ELISA,and the pathological morphology of hippocampus was observed by Nissl staining.Results Compared with normal control group,the expression levels of IL-1βmRNA,TNF-αmRNA and protein in hippocampus of model group were significantly increased(P<0.01,P<0.05);the expression levels of IL-1βmRNA,TNF-αin hippocampus of low-dose group and high-dose group were significantly increased(P<0.01,P<0.05),the expression levels of IL-1βmRNA,TNF-αmRNA and protein in hippocampus of rats in estazolam group were not significantly different(P>0.05).Conclusion Suanzaoren Decoction can reduce the damage of hippocampal nerve in sleep deprived rats by inhibiting neuroinflammation.
